Abstract

Das Myxobakterium Stigmatella aurantiaca bildet unter Hungerbedingungen artspezifische, vielzellige Fruchtkörper. Neben den Genen fbfB und fbfA, deren Produkte in die Fruchtkörperbildung involviert sind, konnten die Gene fbfC und fbfD charakterisiert werden. Diese Gene liegen stromabwärts von fbfA und sind nahe beieinander lokalisiert. Das Startkodon von fbfC überlappt mit dem Stopkodon von fbfA. Die Inaktivierung der Gene bewies die Beteiligung ihrer Genprodukte an der Fruchtkörperbildung. Der putative Promotorbereich von fbfC befindet sich in der kodierenden Sequenz von fbfA. FbfD und fbfC sind in einem Operon organisiert. Die attB site stellt einen Genombereich dar, in den die ortsspezifische Integration von Plasmiden erfolgt, wenn diese die korrespondierende attP site des Myxophagen Mx8 enthalten. Die Integration verändert den Phänotyp, es entstehen Aggregate und pilzartige Strukturen. Die attB site ist in dem 3'-Bereich eines tRNAAsp Gens lokalisiert. Durch die Integration von Plasmiden wird in die stromabwärts-Region des tRNAAsp Gens ein Mx8-spezifischer Terminator eingefügt, der vermutlich für den veränderten Phänotyp verantwortlich ist. Der Phänotyp einer solchen Mutante kann durch eine Integration von Plasmiden über die ortsspezifische Integration in die attR site komplementiert werden, wenn diese Plasmide ein ca. 1.2 kbp großes Fragment der attB site von S. aurantiaca enthalten.

Translation of abstract (English)

The myxobacterium Stigmatella aurantiaca forms species-specific fruiting bodies under starvation conditions. Beside the genes fbfB und fbfA, whose products are involved in fruiting, the genes fbfC and fbfD were characterized. The genes are located downstream of fbfA and are clustered. The start codon of fbfC overlaps with the stop codon of fbfA. The inactivation of the genes showed the involvement of the products in fruiting. The putative promotor region of fbfC is in the coding region of fbfA. FbfD and fbfC are organized in an operon. The attB site is a genome region, in which the site-specific integration of plasmids occurs when they contain the attP site of the myxophage Mx8. The integration causes changes in the phenotype, aggregates and fungus-like structures are visible. The attB site is located in the 3' part of a tRNAAsp gene. The integration of plasmids cause a change in the downstream region of the tRNAAsp gene, a Mx8-specific terminator is inserted that is maybe responsible for the change in the phenotype. The phenotype of such a mutant can be restored by the integration of plasmids by site-specific integration into the attR site These plasmids contain a 1.2 kbp fragment of the attB site of S. aurantiaca.
