Abstract

1)Apparent Dissolution (scheinbare Freisetzung): Untersuchung verschiedener Einflüsse und Entwicklung einer standardisierten Routinemethode. Für die 2007 in der Ph.Eur. monographierte Apparent Dissolution wurden anhand von vier Entwicklungswirkstoffen Einflüsse der Partikelgröße, des Freisetzungsmediums, der Löslichkeit, der Probenbeschickung sowie der Einfluss von Hilfsstoffen in physikalischen Mischungen untersucht. Die Kombination von zwei Probenbeschickungen und einem Freisetzungsmedium mit 0,01 % Tween® 80 sind besonders geeignet für die Charakterisierung der Wirkstoffe. Mit den erhaltenen Daten kann auf die Freisetzung fertiger Formulierungen geschlossen werden. 2)Intrinsic Dissolution (innere Auflösungsgeschwindigkeit): Design eines Systems für Durchflusszellen zur Evaluation neuer Möglichkeiten. Durch die Entwicklung eines Zelleinsatzes zur Intrinsic Dissolution für die monographierte 22,6 mm Tablettenzelle kann erstmals Intrinsic Dissolution in Durchflusszellen durchgeführt werden. Bei einer offenen Betriebsweise kann mittels der entwickelten Kombinationsmethode die Bestimmung der Freisetzungsrate für verschiedene Freisetzungsmedien mittels einer einzigen Freisetzung erfolgen. Verschiedene Metallinlays für den Zelleinsatz ermöglichen die Freisetzung von verschiedenen Presslingsoberflächen. Es wurde ein linearer Zusammenhang zwischen der Oberfläche und der Freisetzungsrate nachgewiesen. Die Anwendung der Kombinationsmethode und die flexible Wahl der Presslingsoberfläche kann die Wirkstoffmenge bei der Bestimmung von Freisetzungsraten erheblich reduzieren. Umwandlungskinetiken, beispielsweise von Salzen an der Presslingsoberfläche, konnten erstmals gemessen und als Kinetik erster Ordnung beschrieben werden. Die Vergleichbarkeit der Freisetzungsraten der Durchflusszelle mit anderen Methoden wurde anhand einer Korrelation mit Freisetzungsraten der häufig verwendeten Distekmethode nachgewiesen. 3)Untersuchung des Einflusses ausgewählter Hilfsstoffe auf die Permeation im Caco-2 Model Die untersuchten Hilfsstoffe Chitosan, Lutrol F68 und emulgierende Stärke zeigten unabhängig von deren Konzentration keinen Einfluss auf die Permeation der beiden Wirkstoffe. Einzig die Auswirkung der Hilfsstoffe auf die Freisetzung kann daher in diesem Fall die Bioverfügbarkeit beeinflussen. Der Einfluss von Hilfsstoffen auf die Permeation in Kombination mit der Apparent Dissolution wird in Zukunft eine immer stärkere Rolle spielen. Da immer neue Hilfsstoffe für eine bessere Freisetzung eingesetzt werden, muss zugleich abgeklärt werden, ob diese Hilfsstoffe eine Auswirkung auf die Permeation und damit die Bioverfügbarkeit haben.

Translation of abstract (English)

1)Apparent Dissolution: Investigation of influences and development of a standardized routine method. The influence of particle size, dissolution medium, solubility, sample loading and excipients in physical mixtures have been investigated since apparent dissolution was observed as monographed in 2007. The combination of two sample loadings and a dissolution medium containing 0.01 % of Tween® 80 is especially suited for the characterisation of active pharmaceutical ingredients (API). According to the recorded data from apparent dissolution experiments it is possible to predict effects on dissolution of API’s in final formulations. 2)Intrinsic Dissolution: Design of a system for flow through cells (FTC) and evaluation of new possibilities and options. Due to the development of an insert for the monographed 22.6 mm tablet cell, intrinsic dissolution has now been realized for the first time in FTC’s. By open loop operation for the developed combined method it is possible to determine the dissolution rate for different dissolution media in one dissolution experiment. The use of different metal inlays for the insert offers flexible dissolution from different surface areas. A linear relationship for different surface areas and dissolution rates was found. Using the combined method and the possibility to select different surface areas, the amount of API could be significantly reduced for the determination of dissolution rates. Conversion kinetics for example of salts at the surface of the compact could be clarified as first order kinetics. With this realisation it may be possible in the future to characterise the conversion of API’s during dissolution without a significant instrumental complexity. The comparability of dissolution rates determined by the FTC with other methods was shown by a correlation to the frequently used distek method. 3)Investigation of the influence of selected excipients on permeation in the caco 2 model. The influence of excipients on permeation as well as the influence of apparent dissolution is important in estimating the impact on bioavailability. The combination of apparent dissolution and the determination of permeation could be crucial when considering the prospective dosage for the formulation, helping to prevent a departure from the therapeutic window. The analysed excipients Chitosan, Lutrol F68 and Purity Gum 2000 show independent of their concentration no influence on permeation of the two observed API’s. Therefore only their influence on dissolution could have an impact on the bioavailability.
