In the last years, the combination of high resolution gel electrophoretic methods and mass spectrometry has been increasingly developed and applied to proteome analysis. Two-dimensional gel electrophoresis (2D) is a powerful and efficient separation technique that enables protein studies from biological material. Fourier transform-ion cyclotron resonance mass spectrometry (FT-ICR-MS) provides high mass resolution, mass accuracy and sensitivity, and thus is an excellent tool for protein structure identification. The combination of two-dimensional gel electrophoresis and FT-ICR mass spectrometry was used in this thesis in biomedical and biochemical applications to proteome analysis, as well for the development of new high resolution mass spectrometric methods.
Zusammenfassung in einer weiteren Sprache
Die Kombination von hochauflösenden gelelektrophoretischen Methoden und Massenspektrometrie wurde in den letzten Jahren mit zunehmendem Erfolg entwickelt und in der Proteomanalytik eingesetzt. Die zweidimensionale (2D) Gelelektrophorese ist eine leistungsfähige und effiziente Methode zur Trennung von komplexen Proteingemischen und ermöglicht die Untersuchung von Proteinstrukturen aus biologischem Material. Die Fourier-Transform-Ionen-Cyclotron-Resonanz-Massenspektrometrie (FT-ICR-MS) ist ein exzellentes Werkzeug zur Proteinidentifizierung, das sich durch hohe Massenauflösung, Massengenauigkeit und Sensitivität auszeichnet. Die Kombination von 2D-Gelelektrophorese und FT-ICR-MS wurde in der vorliegenden Arbeit als analytisches Verfahren in biomedizinischen und biochemischen Proteomuntersuchungen entwickelt und angewendet.
