Zusammenfassung: Iminodisuccinat (IDS) ist ein neuer synthetisch hergestellter Komplexbildner mit dem Potenzial den meistbenutzten aber schwer abbaubaren Komplexbildner Ethylendiamin-tetraacetat (EDTA) teilweise zu ersetzen. Von einem EDTA-Substitut wird neben guten Komplexierungseigenschaften insbesondere eine gute biologische Abbaubarkeit verlangt. In diesem Zusammenhang wurde die Abbaubarkeit von IDS bewertet, die initialen Abbauwege aufgeklärt und die in den Abbau involvierten Gene charakterisiert. Standardisierte aerobe Abbautests nach OECD 301F (Respirometertest) zeigten je nach eingesetztem Belebtschlamm unterschiedliche Ergebnisse. Einerseits wurden mit dem Belebtschlamm aus der Kläranlage Leverkusen-Bürrig alle Isomere des IDS leicht abgebaut. Andererseits wurden nur zwei von drei IDS Isomeren, das R,S-IDS und S,S-IDS, mit dem Belebtschlamm aus der Kläranlage Stuttgart-Büsnau mineralisiert. Des Weiteren wurden die in der Natur hauptsächlich vorliegenden Metall-IDS Komplexe auf ihre biologische Abbaubarkeit untersucht. Dabei erwiesen sich Fe2+- und Ca2+-IDS als biologisch leicht abbaubar, während Mn2+- und Cu2+-IDS innerhalb der 28-tägigen Testperiode nur zu 55% bzw. 40% eliminiert wurden. Aus den genannten Belebtschlämmen wurden die IDS-abbauenden Stämme Agrobacterium tumefaciens BY6 (Leverkusen-Bürrig) und Ralstonia sp. SLRS7 (Stuttgart-Büsnau) isoliert und für weitere Untersuchungen verwendet. In Übereinstimmung mit den Ergebnissen der standardisierten Abbautests mineralisierte der Stamm A. tumefaciens BY6 alle Isomere des IDS, der Stamm R. sp. SLRS7 nur die Isomere R,S-IDS und S,S-IDS. In R. sp. SLRS7 werden R,S-IDS und S,S-IDS über eine homotetramere C-N Lyase reversibel zu Asparaginsäure und Fumarsäure gespalten. Aus R,S-IDS entsteht D-Asparaginsäure, aus S,S-IDS L-Asparaginsäure. Die Spaltung der C-N Bindung erfolgt dabei stereospezifisch am S-konfigurierten C-Atom. In A. tumefaciens BY6 sind zwei Enzyme in den initialen Abbau der IDS Isomere involviert. Zum einen eine C-N Lyase mit vergleichbarer Funktion zur C-N Lyase aus R. sp. SLRS7. Sie katalysiert die stereoselektive Spaltung von R,S-IDS zu D-Asparaginsäure und Fumarsäure. Zum anderen wurde eine Epimerase gefunden. Diese ist das eigentliche Schlüsselenzym für die Metabolisierung von R,R-IDS. Sie epimerisiert R,R-IDS zu R,S-IDS sowie S,S-IDS zu R,S-IDS. Hierdurch werden die Isomere R,R-IDS und S,S-IDS in den stereoselektiven Pfad eingeleitet, der durch die C-N Lyase katalysiert wird. Die Gene für die Epimerase und die C-N Lyase aus A. tumefaciens BY6 wurden kloniert und sequenziert. Die von dem Lyase-Gen abgeleitete Aminosäuresequenz zeigte die höchste Homologie mit Argininosuccinat-Lyasen, einer Unterfamilie der Fumarase II Superfamilie. Eine entsprechende Lyase-Aktivität gegenüber Arginiosuccinat wurde jedoch nicht beobachtet. Bei der C-N Lyase aus A. tumefaciens BY6 handelt es sich um ein nahe verwandtes Enzym der Argininosuccinat-Lyasen mit bisher unbekannter Hauptfunktion. Die von dem Epimerase-Gen abgeleitete Aminosäure-sequenz besaß keine signifikante Ähnlichkeit zu Proteinen mit bekannter Funktion. Die Epimerase kann demnach als neuartig bezeichnet werden. Die beiden in den IDS-Abbau involvierten Gene zeigten keine funktionelle Kopplung. Das Epimerase-Gen wurde auf einem zirkulären Plasmid, das Lyase-Gen auf der chromosomalen DNA lokalisiert.
Iminodisuccinate (IDS) is a new synthetic chelating agent with the potential to substitute partially the most commonly used but persistent chelator ethylenediaminetetraacetate (EDTA). An EDTA-substitute requires besides good complexing attributes in particular ready biodegrability. In this regard the biodegradability of IDS was estimated, the initial degradation pathway was elucidated, and the genes involved in the IDS-degradation were characterized. The standardized aerobic biodegradation tests revealed different results depending on the activated sludge used. On the one hand, using the activated slugde from the sewage treatment plant of Leverkusen-Bürrig all isomers of IDS were readily biodegraded. On the other hand, only two of three IDS-isomers, namely R,S-IDS and S,S-IDS, were mineralized by the activated sludge from the sewage treatment plant of Stuttgart-Büsnau. Furthermore, in natural systems the mostly occuring metal-IDS complexes were tested on its biodegradability. Fe2+- and Ca2+-IDS were readily biodegradable whereas Mn2+- und Cu2+-IDS revealed only 55% and 40% biodegradation, respectively, within the 28 day period of the test. From the activated sludges mentioned the IDS-degrading strains Agrobacterium tumefaciens BY6 (Leverkusen-Bürrig) and Ralstonia sp. SLRS7 (Stuttgart-Büsnau) were isolated and used for further investigations. In agreement with the results of the standardized biodegradation tests, the strain A. tumefaciens BY6 mineralized all isomers of IDS, the strain R. sp. SLRS7 only the isomers R,S-IDS and S,S-IDS. The strain R. sp. SLRS7 cleaved R,S-IDS and S,S-IDS by a homotetrame C-N lyase with formation of aspartic acid and fumaric acid. Actually, R,S-IDS and S,S-IDS led to D-aspartic acid and L-aspartic acid, respectively. The cleavage of the C-N bond takes place stereospecifically on the asymmetric carbon atom exhibiting the S-configuration. In contrast, two enzymes are involved in the initial transformation of all IDS isomers by A. tumefaciens BY6. There is a C-N lyase with comparable function to the C-N lyase from R. sp. SLRS7. It catalyzes the stereoselective cleavage of R,S-IDS generating D-aspartic acid and fumaric acid. However, the key enzyme for R,R-IDS metabolism is an epimerase. It epimerizes R,R-IDS to R,S-IDS as well as S,S-IDS to R,S-IDS. Hereby, the isomers R,R-IDS and S,S-IDS were funneled into the stereoselective pathway initiated by the C-N lyase. The genes encoding the epimerase and the C-N lyase were cloned and sequenced. The deduced amino acid sequence of the C-N lyase gene showed the highest degree of sequence similarity with argininosuccinate lyases, a subfamilie of the fumarase II superfamily. However, an appropriate lyase activity towards argininosuccinate could not be observed. The C-N lyase from A. tumefaciens BY6 is a closely related enzyme to the argininosuccinate lyase and so far with unknown natural function. The deduced amino acid sequence of the epimerase gene exhibited no significant similarity to proteins with known function. Therefore, the epimerase is described as a novel enzyme. Both genes involved in IDS degradation showed no functional coupling. The epimerase gene was located on a circular plasmid, the lyase gene was part of the chromosomal DNA.
