Zusammenfassung: Bei der Herstellung von Antibiotika werden in großem Umfang Bakterien eingesetzt. Dabei handelt es sich vor allem um Bakterien der Gattung Actinomyceten. Daher sind diese Organismen heute in der Biotechnologie von großer Bedeutung. Actinomyceten kommen im Gegensatz zu anderen Bakterien nicht als Einzeller vor, sondern bilden ein fadenförmiges Geflecht, ein Myzel, aus. Dieses kann als Substrat- oder Luftmyzel ausgeprägt sein. Bei ihren Produkten handelt es sich meist um Sekundärstoffe mit antibiotischer Wirkung, welche für das Wachstum der Organismen nicht notwendig sind. Die Regulation des Sekundärstoffwechsels von Actinomyceten ist sehr komplex und es sind noch nicht alle Einzelheiten der ablaufenden Stoffwechselvorgänge und vor allem deren Regulation aufgeklärt. Um eine Optimierung des Herstellungsprozesses zu erreichen, ist es sinnvoll, heutige Methoden der Systemtheorie anzuwenden. Eine Voraussetzung ist dabei die Erstellung eines mathematischen Modells zur Beschreibung des Wachstums- und Produktbildungsverhaltens der Mikroorganismen. Dieses Modell kann dann eingesetzt werden, um eine optimierte Prozeßführung durchzuführen. Die Erstellung und Anpassung eines solchen mathematischen Modells für einen bestimmten Stamm kann sehr zeitaufwendig sein. Daher ist es wünschenswert, ein möglichst allgemeingültiges Modell zu entwickeln, welches schnell auf andere Stämme und Produkte angepaßt werden kann. In der vorliegenden Arbeit wird auf der Basis bereits vorhandener experimenteller Ergebnisse und Modelle für das Wachstumsverhalten, sowie eigener experimenteller Ergebnisse ein verallgemeinertes, strukturiertes mathematisches Modell erstellt. Die Modellparameter werden an die beiden untersuchten Actinomyceten Stämme Streptomyces tendae und Amycolatopsis mediterranei angepaßt. Damit wird die Grundlage geschaffen, um bei verschiedenen Stämmen von Actinomyceten eine modellgestützte Optimierung der Prozeßführung zu erreichen.
A large variety of bacteria are currently used for the production of antibiotics. Most of the bacteria are of the species Actinomyces. Therefore, Actinomyces play an important role in biotechnology today. Since they are mycelial organisms, their morphology is more complex than the morphology of single cell organisms. The mycelia can be divided into substrate and airial mycelia. Their products are mostly secondary metabolites with antibiotic effects, they are not necessary for their own growth. The regulation of the secondary metabolism is very complex and some of the details of the metabolic pathways and especially their regulation are still unknown. System theory including process modelling can be used in order to optimize the manufacturing process. Therefore, one prerequisite is the existance of a mathematical model for the description of the growth and product formation of the organism. The way to obtain such a model can be very time consuming. This clearly indicates the need for a general model description which can be used for different strains and products, only by changing the model parameters. In the present work, a general structured mathematical model is obtained on the basis of already available experimental results and a model for the growth, as well as new experimental results which are obtained during the work for the thesis. The model parameters are fitted for the two examined actinomyces strains Streptomyces tendae and Amycolatopsis mediterranei. On the basis of the resulting model a process optimization of the production is possible.
