Abstract
  1. Background and Aims There are studies showing a correlation between interleukin-1β (IL-1β), interleukin-6 (IL-6) and the activity of the acid sphingomyelinase (ASM). Moreover a positive correlation between IL-1β concentration and the activity of the ASM has been described. Further studies showed, that IL-1β affects IL-6 positively, IL-6 in contrast inhibits IL-1β. The aim of this dissertation was to investigate a hypothetical correlation between the cellular (lysosomal) ASM activity in human peripheral blood mononuclear cells (PBMCs) and the quantitative concentration of both interleukins. Therefore venous blood samples of 156 probands were analyzed. In addition the influence of age and sex on the concentration of the interleukins was examined, as well as the effect of sex on the ASM activity. Also the correlation between the concentrations of the interleukins was examined. 2) Methods (Probands, Materials and Quantification techniques) To measure the concentration of IL-1β and IL-6 as well as the activity of the ASM, venous blood samples were taken from the test persons. Both interleukins were measured in the plasma of the probands using ELISA technique. Data of the ASM activity already existed. The ASM activity was determined in mononuclear leukocytes, which were isolated using density gradient centrifugation. 3) Results and Observations The ASM activity values, measured at 156 test persons, showed a Gaussian distribution. One value was not measurable. The average ASM values were 1,85 ± 0,62 nmol/mg/h (mean value ± standard deviation; n=155). The IL-6 values, measured at 137 participants, did not show Gaussian distribution. The average IL-6-Konzentration was 2159,05 ± 2677,14 fg/ml (mean value ± standard deviation; n=137). Also the IL-1β-concentration, measured at 125 test persons, did not show Gaussian distribution. The exact data were 737,55 ± 989,14 fg/ml (mean value ± standard deviation; n=125). No significant correlations were seen between the IL-6- respectively IL-1β-concentration and the activity of the ASM in this sample (p=0,972 and p=0,300). Furthermore no significant influence of age on the interleukin concentration was apparent (IL-6: p=0,151 and IL-1β: p=0,214) and also the consideration of sex being related to the interleukins (IL-6: p=0,925 and IL-1β: p=0,438) as well as being related to the ASM-activity (p=0,599) did not show significant differences. The IL-6- and the IL-1β-concentrations showed a highly significant positive correlation (p=0,001). 4) Conclusions A larger number of test persons could possibly minimize the effects of deviation in the ASM-activity and the interleukin concentrations and in the following a correlation between interleukins and ASM might be detected. Also a consideration of chronically ill patients, who generally show higher levels of interleukin concentration, could enable the detection of the expected but not found correlation.

  1. Hintergrund und Ziele Studien weisen auf einen Zusammenhang zwischen Interleukin-1β (IL-1β), Interleukin-6 (IL-6) und der Aktivität der sauren Sphingomyelinase (ASM) hin. So konnten diese eine positive Beziehung zwischen IL-1β-Konzentration und ASM-Aktivität zeigen. Man weiß zudem, dass IL-1β IL-6 induziert, IL-6 jedoch wiederum auf IL-1β hemmend wirkt. Ziel dieser Dissertation war es den Zusammenhang zwischen der Aktivität der zellulären (lysosomalen) ASM in menschlichen PBMCs (peripheral blood mononuclear cells) und der quantitativen Konzentration der beiden genannten Interleukine zu untersuchen. Hierzu wurde das venöse Blut von 156 Probanden untersucht. Der Einfluss von Alter und Geschlecht auf die Konzentrationen der Interleukine wurde betrachtet, sowie der Einfluss des Geschlechts auf die Aktivität der ASM. Auch die Korrelation der Konzentrationen der Interleukine untereinander wurde untersucht. 2) Methoden (Probanden, Material und Untersuchungsmethoden) Zur Bestimmung der Konzentration von IL-1β und IL-6 sowie der Aktivität der ASM wurde den Probanden venöses Blut entnommen. Die beiden Interleukine wurden im Plasma der Probanden mittels ELISA-Technik bestimmt. Die Daten über die Aktivität der ASM waren am untersuchten Kollektiv bereits vorhanden. Die ASM-Aktivität wurde in mononukleären Leukozyten, welche mittels Dichtegradientenzentrifugation isoliert wurden, bestimmt. 3) Ergebnisse und Beobachtungen Die ASM-Aktivitätswerte, die bei 156 Probanden gemessen wurden, zeigten im Histogramm eine Normalverteilung. Bis auf einen Wert waren alle Aktivitäten messbar. Die Werte lagen im Mittel bei 1,85 ± 0,62 nmol/mg/h (Mittelwert ± Standardabweichung; n=155). Die bei 137 Probanden gemessenen IL-6-Werte folgten keiner Normalverteilung. Im Mittel lag die IL-6-Konzentration bei 2159,05 ± 2677,14 fg/ml (Mittelwert ± Standardabweichung; n=137). Auch die IL-1β-Konzentrationen, die bei 125 Probanden gemessen wurden, wiesen keine Normalverteilung auf. Die Werte lagen bei 737,55 ± 989,14 fg/ml (Mittelwert ± Standardabweichung; n=125). Es wurden keine signifikanten Zusammenhänge zwischen der IL-6- bzw. IL-1β-Konzentration und der Aktivität der ASM in dieser Stichprobe beobachtet (p=0,972 bzw. p=0,300). Desweiteren wurde kein signifikanter Einfluss des Alters auf die Interleukinkonzentrationen ersichtlich (IL-6: p=0,151 bzw. IL-1β: p=0,214) und auch bei der Betrachtung des Geschlechts in Bezug auf die Interleukine (IL-6: p=0,925 bzw. IL-1β: p=0,438), sowie in Bezug auf die ASM-Aktivität (p=0,599), ergaben sich keine signifikanten Unterschiede. Die IL-6- und die IL-1β-Konzentrationen korrelierten hoch signifikant positiv miteinander (p=0,001). 4) Praktische Schlussfolgerungen Möglicherweise könnte man durch eine größere Probandenzahl die Auswirkung der Schwankungen in der ASM-Aktivität und den Interleukin-Konzentrationen minimieren und eine Korrelation zwischen den Interleukinen und der ASM nachweisen. Auch könnte eine Betrachtung von chronisch kranken Patienten, die in der Regel höhere Interleukin-Konzentrationen aufweisen, den Nachweis dieser Korrelation ermöglichen.
