Abstract

Background and objectives During placentogenesis several fusion events contribute to the formation of the sycytiotrophoblast. Trophoblast fusion is characterized by the increased gap junction connexin abundance, and additionally by the reduction of desmoplakins accompanied by disintegration of the cell membrane. It is assumed that these processes are amongst others initiated and regulated by the protein syncytin. The objective of this study was to visualise cell fusion events using a cell fusion assay with trophoblasts and syncytin overexpressing cells. Methods In this study we analyzed cellular effects of an increased or reduced syncytin expression in genuine trophoblasts, stably syncytin transfected cells (CHO/52), trophoblast cell lines (BeWo) and with a control cell line (CHO) by using a fluorescent cell fusion assay, immunocytochemical connexin 43 and desmoplakin stainings. An increased syncytin expression was achieved by different stimulants (Forskolin, 8-BrcAMP, supernatant from syncytin transfected CHO/52 cells, CaCl2). Results and observations Using the fluorescent cell fusion assay it was demonstrated that the fusion rate of genuine trophoblast cells, stably syncytin transfected cells and trophoblast cell lines (BeWo) as well as connexin 43 immunoreactivity in these cells increased with incubation time and by adding stimulants, which was in contrast to the control cell line. Simultaneously, a reduction of desmoplakin immunoreactivity was observed in genuine trophoblast cells, stably syncytin transfected cells and trophoblast cell lines (BeWo). Conclusions Our results confirm the fusiogenic properties of syncytin. Using a fluorescent cell fusion assay changes in in vitro syncytialisation rates could be demonstrated. These changes were accompanied by corresponding alterations in the abundance of gap junction and desmosomal proteins. During pregnancy, syncytin is essential for placental function. In pregnancy-associated disorders such as preeclampsia a disturbed gene expression or altered protein function has been discovered. We assume that hypoxia is a risk factor for a disturbed syncytialisation because of down-regulation of syncytin gene expression. To elucidate the function of syncytin in detail, further research is necessary in the context of physiological and pathophysiological placental development.

Abstract

Hintergrund und Ziele Während der Plazentogenese sind mehrere Fusionsvorgänge an der Bildung des Syncytiotrophoblasten beteiligt. Sichtbar ist die Fusion unter anderem durch die vermehrte Ausbildung von Connexinen den gap junctions, und die Reduktion von Desmoplakinen im Rahmen der Auflösung der Zellmembran. Es wird angenommen, dass diese Vorgänge unter anderem vom Protein Syncytin initiiert und reguliert werden. Ziel der Studie war es die Zellfusionsvorgänge durch einen Fluoreszenz-Fusionsassay an trophoblastären und Syncytin-überexprimierenden Zellen zu visualisieren. Methoden In dieser Arbeit wurde an genuinen Trophoblasten, stabil Syncytin transfizierten Zellen(CHO/52), an einer trophoblastären Zelllinie (BeWo) sowie an einer Kontroll-Zellline(CHO) mit Hilfe eines Fluoreszenz-Zellfusionsassays, Connexin 43 und Desmoplakin Färbungen qualitativ die zellulären Effekte einer erhöhten oder erniedrigten Syncytinexpression untersucht. Eine erhöhte Syncytinexpression wurde durch verschiedene Stimulantien (Forskolin, 8-Br- cAMP, Mediumüberstand aus Syncytin transfizierten CHO/52 Zellen, CaCl2) erreicht. Ergebnisse und Beobachtungen Es konnte mittels des Zellfusionsassays gezeigt werden, dass sowohl die Fusionsrate genuiner trophoblastärer Zellen, Synyctin transfizierter Zellen und trophoblastärer Zelllinien (BeWo) als auch die Connexin 43 Immunreaktivität im Gegensatz zur Kontroll-Zelllinie mit der Inkubationsdauer und unter Stimulantienzugabe zunahm. Parallel dazu kam es zu einer Reduktion der Desmoplakin Immunreaktivität in genuinen trophoblastären Zellen, Synyctin transfizierten Zellen und trophoblastären Zelllinien (BeWo). Praktische Schlussfolgerungen Unsere Ergebnisse bestätigen, dass Syncytin fusiogene Eigenschaften hat. Mittels des genutzten Fusionsassays konnten Veränderungen in der Syncytialisierungsrate in vitro dargestellt werden. Diese gingen einher mit entsprechenden Veränderungen in der Nachweisbarkeit von gap junction und Desmosomen Proteinen. Syncytin ist während der Schwangerschaft für die Plazentafunktion essentiell. Bei Schwangerschaftsassoziierten Erkrankungen wie z.B. der Präeklampsie konnte eine gestörte Gen- oder Proteinexpression gezeigt werden. Es wird angenommen, dass u.a. Hypoxie durch Hemmung der Syncytin Expression auf transkriptioneller Ebene ein Risikofaktor für eine gestörte Syncytialisierung ist. Um die Rolle von Syncytin weiter zu verstehen, müssen weitere Untersuchungen im Rahmen der physiologischen und pathologischen plazentaren Entwicklung folgen.
