Zusammenfassung: 
Pradofloxacin – ein neues C8-Cyanofluorchinolon – wurde für die Behandlung bakterieller Infektionen im veterinärmedizinischen Bereich entwickelt. In dieser Arbeit wurde die Aktivität von Pradofloxacin auf pro- und eukaryontische Typ II-Topoisomerasen im Vergleich zu anderen Fluorchinolonen neben der Bestimmung der MHK als „klassischer“ Methode mit Hilfe von Absterbekinetiken, Selektionsversuchen, Cleavage-Assays und einem neuentwickelten In-vivo-Testsystem bestimmt.

Pradofloxacin weist im Vergleich zu Enro-, Marbo-, Cipro- und Moxifloxacin eine höhere antibakterielle Wirkung gegenüber Wildtypstämmen und resistenten Typ II-Topoisomerasemutanten von Escherichia coli, Staphylococcus aureus und Staphylococcus intermedius auf. Die Substanz ist darüber hinaus aktiv gegenüber hochresistenten Stämmen von E. coli (mit genetisch definierten Mutationen in den Genen der Zielstrukturen und der Regulatoren von Effluxpumpen sowie einer verminderten Porinbildung).
Als bevorzugte Zielstruktur von Pradofloxacin in E. coli wurde Gyrase bzw. in S. intermedius Topoisomerase IV identifiziert.
Pradofloxacin besitzt ein geringes Potential zur Selektion resistenter Bakterien und ist aktiv gegenüber Zellen von E. coli, S. aureus und S. intermedius Wildtyp- und Mutantenstämmen in verschiedenen physiologischen Stadien. Beides kann dazu beitragen, eine Resistenzentwicklung während einer Therapie zu verhindern bzw. zu verlangsamen.
In Wildtyp- und isogenen Mutantenstämmen von E. coli erfolgte die Einleitung des bakteriellen Zelltods – in Folge von DNA-Doppelstrangbrüchen – durch Pradofloxacin im Gegensatz zu Enro-, Marbo-, Cipro- und Moxifloxacin bereits in Anwesenheit geringerer, subinhibitorischer Konzentrationen. Die Fähigkeit von Pradofloxacin, bakterielle Zellen bereits bei sehr geringen Konzentrationen zu schädigen, stellt eine mögliche Erklärung für die herausragende antibakterielle Wirkung dieser Substanz dar.
Für die antibakterielle Wirkung von Pradofloxacin spielen die C8-Cyanogruppe und der C7-Pyrrolopiperidinring vermutlich eine wesentliche Rolle. Beide Substituenten scheinen sowohl für die Aktivität der Substanz auf die bakteriellen Zielstrukturen als auch für eine verringerte Affinität zu Effluxpumpen bzw. eine erleichterte Passage der äußeren Membran gramnegativer Bakterien von Bedeutung zu sein.
Im Vergleich zu humaner Topoisomerase II besitzt Pradofloxacin eine hohe Selektivität für E. coli Gyrase (800-fach). Bei Einsatz physiologischer Konzentrationen von Pradofloxacin während der Therapie bakterieller Infektionen ist eine Hemmung von eukaryontischen Topoisomerasen somit nicht zu erwarten.

Das in dieser Arbeit neu entwickelte In-vivo-Testsystem (precA-luc-Reportergenassay) erlaubt eine empfindliche Detektion von Substanzkonzentrationen, die zur Induktion der SOS-Antwort führen. Es kann somit für ein sensitives Screening nach Substanzen eingesetzt werden, die über eine Hemmung der Replikation oder auch eine direkte Wechselwirkung mit der DNA die SOS-Antwort induzieren. Darüber hinaus eignet sich das Testsystem zur Ermittlung der antibakteriellen Restaktivität von Fluorchinolonmetaboliten. Dies konnte am Beispiel von Enrofloxacin-N-4’-oxid demonstriert werden. Die Ermittlung der antibakteriellen Restaktivität erscheint sinnvoll, da durch Fluorchinolonmetaboliten mit einer antibakteriellen Restaktivität theoretisch die Selektion Fluorchinolon resistenter Bakterien möglich ist.

Pradofloxacin – a new C8-cyanofluoroquinolone – has been developed for the treatment of bacterial infections in veterinary medicine. In this study the activity of pradofloxacin against both, pro- and eukaryotic typ II topoisomerases was determined by MIC determination as “conventional” method, kill-kinetics, the ability to select resistant mutants, cleavage-assays, and a new developed in-vivo-test system compared to other fluoroquinolones.

Pradofloxacin efficiently kills wildtyp strains and resistant typ II topoisomerase mutants of Escherichia coli, Staphylococcus aureus and Staphylococcus intermedius compared to enro-, marbo-, cipro- und moxifloxacin. In addition, pradofloxacin is active against high-level resistant mutants of E. coli with genetically defined mutations in the genes of the target enzymes and different regulators of efflux pumps and decreased expression of porins.
Gyrase and topoisomerase IV were identified as primary targets of pradofloxacin in E. coli, and S. intermedius, respectively.
Pradofloxacin posses a low potential to select resistant mutants and is able to kill cells of E. coli, S. aureus und S. intermedius wildtyp and mutant strains in different physiological stages. These properties could help to prevent or at least minimize the development of resistant mutants during therapy.
In wildtyp and resistant isogenic mutants of E. coli, bacterial cell death – as a result of DNA double strand breaks – occurs at concentrations of pradofloxacin much lower than those determined for enro-, marbo-, cipro- and moxifloxacin and far below the MIC. The ability of pradofloxacin to injure bacterial cells at lowest concentrations convincingly explains the superior antibacterial activity of the drug.
Probably, the C8-cyano- und C7-pyrrolopiperidin substituents play a critical role for the antibacterial activity of pradofloxacin. Both substituents seem to be important for the high activity against the bacterial target enzymes as well as the reduced affinity for efflux pumps and permeability of the outer membrane of gramnegative bacteria.
Pradofloxacin is highly selective for E. coli gyrase (800-fold) compared to human topoisomerase II. During treatment of bacterial infections with physiological concentrations of pradofloxacin inhibition of eukaryotic topoisomerases should not occur.

The newly developed in-vivo-test system (precA-luc reporter gene assay) allows for a highly sensitive detection of those drug concentrations leading to the induction of the SOS regulon. It can be used to screen for compounds which induce the SOS-response due to either an inhibition of the DNA replication or a direct interaction with DNA. Moreover, the in-vivo-test system is suitable to investigate the residual antibacterial activity of fluoroquinolone metabolites. This could be demonstrated with enrofloxacin-N-4’-oxide as an example for fluoroquinolone metabolites with a residual antibacterial activity which could select fluoroquinolone resistant bacteria in the environment. So it seems to be efficient to determine the residual antibacterial activity of fluoroquinolone metabolites.
