Die Erkenntnis, dass ATP und NAD neben ihrer intrazellulären Rolle im Energiestoffwechsel auch extrazellulär eine Rolle als Signalmoleküle spielen, ist relativ neu und noch nicht ausgiebig untersucht. Beide Nukleotide aktivieren auf unterschiedliche Art den Purinrezeptor P2X7, der auf vielen hämatopoetischen Zellen exprimiert wird und an der Regulation von Entzündungsvorgängen beteiligt ist. Der P2X7-Rezeptor wird entweder direkt durch Bindung von ATP aktiviert, oder indirekt durch eine ADP-Ribosyltransferase (ART), die NAD nutzt um den Rezeptor zu modifizieren (ADP-ribosylieren). Die Aktivierung des P2X7-Rezeptors führt u. a. zu Veränderungen der Zellmembran (z. B. Externalisierung von Phosphatidylserin) und zur Prozessierung und Sekretion von Interleukin-(IL-)1b. In dieser Arbeit wurden die Wirkungen von NAD und ATP auf antigenpräsentierende Zellen (APZs) und T-Zellen untersucht.
Neben der Expression wurde die Funktion des P2X7 auf APZs und T-Zellen anhand einer Reihe messbarer, durch die Aktivierung des Rezeptors ausgelösten Veränderungen analysiert. Dabei zeigten die T-Zellen, abgesehen von der IL-1b-Sekretion, eine deutlich stärkere Reaktion als die APZs, obwohl die Oberflächenexpression des Rezeptors auf beiden Zellarten vergleichbar war.
Sowohl Makrophagen als auch dendritische Zellen werden durch den Kontakt mit T Zellen in Gegenwart von Antigen zur Sekretion von IL-1b angeregt. Diese antigenabhängige IL-1b Sekretion erwies sich in beiden Zellarten als P2X7-unabhängig. Dagegen zeigten Makrophagen und dendritische Zellen Unterschiede in der antigenunabhängigen, durch LPS induzierten IL-1b-Sekretion. Diese war in Makrophagen P2X7-abhängig, in dendritischen Zellen jedoch P2X7-unabhängig. Die Unterschiede im Mechanismus der IL-1b Sekretion weisen auf eine unterschiedliche Rolle von P2X7 in diesen beiden Typen von APZs gemäß ihrer unterschiedlichen Rolle in der Immun¬antwort hin.
Die Aktivierung von P2X7 durch NAD ist von der Gegenwart einer ART abhängig. Im Kontext des Immunsystems wurden bisher nur die Isoformen ART2.1 und ART2.2 aus¬schließlich auf T-Zellen nachgewiesen. In dieser Arbeit wurde nun erstmals gezeigt, dass ART2.1 auch auf verschiedenen Populationen von APZs exprimiert wird und enzymatisch aktiv ist. Die Expression von ART2.1 wurde auf Makrophagen durch Stimulation mit LPS oder Interferonen gesteigert.
Die ADP-Ribosylierung des P2X7-Rezeptors durch ART2.1 auf Makrophagen führte nicht zu einer vollständigen Aktivierung des Rezeptors, wie dies bei der ART2.2-vermittelten ADP-Ribosylierung von P2X7 auf T-Zellen der Fall ist, sondern senkte die Schwelle für die Aktivierung von P2X7 durch ATP. Schließlich wurden neben dem P2X7-Rezeptor beide Ketten der Integrine LFA-1 (CD11a/CD18) und CR3 (CD11b/CD18) als Zielproteine der ART2.1 auf Makrophagen identifiziert.

The nucleotides NAD and ATP play a major role in the intracellular energy metabolism of cells. The knowledge of their function as extracellular signalling molecules is relatively new and far from being exhaustively studied. Both nucleotides activate, via different means, the P2X7 purinergic receptor, which is expressed on a wide range of hematopoietic cells and participates in the regulation of inflammatory responses. The P2X7 receptor is activated either directly by binding its ligand ATP or indirectly by an ADP-ribosyltransferase (ART) using NAD as a substrate to modify (ADP-ribosylate) the receptor. Activation of the P2X7 receptor leads amongst others to changes in the plasma membrane (i. e. externalisation of phosphatidylserine) and to the processing and secretion of interleukin-(IL-)1b. In the course of this study the effects of NAD and ATP on antigen-presenting cells (APCs) and T cells were examined.
Experiments examining the expression and function of the P2X7 receptor on APCs and T cells showed a stronger reaction of T cells than APCs in most assays of P2X7 receptor activation, except for the secretion of IL-1b, although P2X7 surface expression on the two cell types was comparable.
Both macrophages and dendritic cells (DCs) secreted IL-1b upon interaction with T cells in the presence of antigen. This antigen-dependent IL-1b secretion proved to be independent of the P2X7 receptor. In contrast there were differences between macrophages and DCs regarding the antigen-independent IL-1b secretion induced by incubation with LPS. While the antigen-independent IL-1b secretion was dependent on P2X7 in macrophages, it was independent of P2X7 in DCs. Differences in the mechanism of IL-1b secretion point to a different role of the P2X7 receptor on these two types of APCs according to their different roles in the course of an immune response.
Activation of the P2X7 receptor by NAD depends on the presence of an ART. In the context of the immune system, only the ART2 isoforms ART2.1 and ART2.2 have been described so far, and their expression is thought to be restricted to T cells. In this study it was shown for the first time that ART2.1 is also expressed and enzymatically active on different populations of APCs. On macrophages the expression of ART2.1 on macrophages was increased upon stimulation with LPS or interferones.
ADP-ribosylation of the P2X7 receptor on T-cells by ART2.2 leads to full activation of the receptor. In contrast NAD-dependent ADP-ribosylation of the P2X7 receptor on macrophages by ART2.1 does not activate the receptor by itself, but lowers the threshold for ATP-mediated activation. Finally, in addition to the P2X7 receptor, both chains of the integrins LFA-1 (CD11a/CD18) and CR3 (CD11b/CD18) could be identified as novel target proteins of ART2.1 on macrophages.
