Ziel der vorliegenden Arbeit war, supramolekulare Eigenschaften des [µ-Chloro-bis{chloro-(cyclopentadienyl)-methyl-4,6,-O-benzyliden-b-D-glucopyranosidato-1kO2,1:2kO3-zirconat}]-Anions (1–) zu erforschen, das bei der Umsetzung von CpZrCl3(thf)2 mit Methyl-4,6-O-benzyliden-b-D-glucopyranosid im Gegenwart von Triethylamin in guten Ausbeuten gebildet wird. Das komplexe Anion 1– verfügt über eine C2-symmetrische molekulare Struktur in Lösung und bindet ein Ammonium-Kation endohedral in seiner chiralen Tasche über eine Wasserstoffbrücke.
Die Bindung des Triethylammonium-Gastes an den Zirconat-Wirt 1– in Lösung konnte in dieser Arbeit durch ein 1H-DOSY-Experiment nachgewiesen werden. Ferner konnte gezeigt werden, dass in Lösung nahezu beliebige primäre, sekundäre und tertiäre Amine mit dem Triethylamin in der Kavität von 1– schnell austauschen, je nach sterischem Anspruch und Basizität des neuen Amins. Die prinzipielle, auf der Reaktionsenthalpie basierende Austauschmöglichkeit einiger Amine in der Tasche von 1– wurde mit Hilfe der DFT-Methode berechnet. Die Ergebnisse dieser Rechnungen stimmen qualitativ mit den experimentellen Befunden überein.
Optisch aktive, als Racemat eingesetzte Amine tauschen ebenfalls gegen Triethylamin in der chiralen Kavität von 1– aus. Durch diastereotope Wechselwirkungen des chiralen Wirtes mit dem Gast wird eine eindeutige Zuordnung der 1H-NMR-Signale zu den entsprechenden Enantiomeren der Amine möglich. Damit kann das Zirconat-Anion 1– zur chiralen Erkennung eingesetzt werden.
1H-NMR-Experimente zeigen ferner, dass die Bindungsenergie des (S)-Enantiomers von 1-Phenylethylamin mit der Tasche von 1– größer als die des (R)-Enantiomers ist. DFT-Rechnungen zu diesen Aspekt konnten diesen Befund bestätigen. Damit lässt sich die chirale Erkennung präparativ nutzen, wie bei der Amidierung eines Enantiomerengemisches des Amins mit Pivaloylchlorid gezeigt wurde. Im Experiment wurde ein Enantiomerenüberschuss von 33% für das endohedral gebundene (S)-Enantiomer nachgewiesen.
Der Versuch, Enamine als Dienophile für Diels-Alder-Reaktionen durch Protonierung des Stickstoffatoms in der chiralen Tasche von 1– zu aktivieren, führte zur Bildung eingeschlos-sener Iminium-Kationen. Iminium-Komplexe des Zirconat-Anions 1– sind durch die endohedrale Bindung so stabilisiert, so dass sie sich als entsprechende Wirt-Gast-Komplexe isolieren lassen. Im Gegensatz dazu wird ohne Anwesenheit des Zirconat-Anions 1– nur eine Gleichgewichtsreaktion zwischen freiem Enamin und Triethylammoniumchlorid beobachtet. Dann enthielten Lösungen sowohl das Enamin als auch das Iminium-Kation. Beim Einengen solcher Lösungen zur Trockne wird das Enamin quantitativ zurückgewonnen.
Die Bildung der Iminium-Kationen innerhalb der chiralen Tasche von 1– öffnet im Prinzip Wege zur enantioselektiven nukleophilen Addition an das a-Kohlenstoffatom des Iminium-Kations. Während der Versuch der Addition von Kohlenstoff-Nucleophilen wie Malonat und Benzoylacetonat nicht erfolgreich war, verlief die Addition von Cyanid sowohl mit Triethylammoniumchlorid als auch mit dem Komplex [Et3NH]1 in sehr guten Ausbeuten zu den a-Aminonitrilen, die Vorstufe zu a-Aminosäuren sind.
Ein stereochemischer Einfluss des Zirconat-Anions 1– auf die Bildung des a-Aminonitrils wurde für 1-(2-Phenyl-1-propenyl)-pyrrolidin beobachtet, das nach der Transformation zu a-Aminonitril zwei stereogene Zentren enthält und in Form der erythro- und threo-Diastereomeren existiert. Die modifizierte Strecker-Reaktion in Anwesenheit des Zirconat-Komplexes [Et3NH]1 erhöht gegenüber der Reaktion mit HNEt3Cl den Anteil des threo-Diastereomers im Vergleich zu erythro-Isomers um das 7.5-fache. Dieser Effekt ist offensichtlich eine Folge der stereochemischen Induktion der chiralen Kavität von 1–.
Die Untersuchungen der Eigenschaften der chiralen Tasche von 1– zeigten nicht nur, dass das endohedral gebundene Triethylamin gegen andere Amine und Enamine ausgetauscht wird, sondern dass diese Amine und Enamine auch zur direkten Synthese von 1– als Abfang-reagenzien des gebildeten Chlorwasserstoffs eingesetzt werden können. Die gebildeten supramolekularen Komplexe enthalten die neuen Amine und Enamine als Ammonium- und Iminium-Kationen in der Tasche von 1–. Da die chirale Tasche von 1– Enantiomere optisch aktive Amine unterscheidet, kann in der Synthese mit dem racemischen Gemisch eines chiralen Amins ein Enantiomer bevorzugt in die Kavität von 1– eingebaut werden. Mit diesem Verfahren wurde für das in die Tasche von 1– eingelagerte 1-Phenylethylamin ein beachtlicher Enantiomerenüberschuss des (S)-Enantiomers von 65% erhalten.

The objective of this work was to investigate the supramolecular properties of the [µ-chloro-bis{chloro-(cyclopentadienyl)-methyl-4,6,-O-benzylidene-b-D-glucopyranosidato-1kO2,1:2kO3-zirconate}]-anion (1–), which is formed in good yields by reaction of CpZrCl3(thf)2 with methyl-4,6-O-benzylidene-b-D-glucopyranoside in the presence of triethylamine. The complex anion 1– has a C2-symmetric molecular structure in solution and binds an ammonium cation endohedrally in its chiral pocket via the hydrogen bridge. The binding of the triethylammonium guest in the host 1– was demonstrated by the 1H DOSY experiment.
In addition it was shown that in solution almost any primary, secondary and tertiary amines rapidly exchange with triethyl-amine in the cavity of 1–, depending on the steric demands and basicity of the amine. Based on the reaction enthalpy the possibility of exchange for some amines in the pocket of 1– was calculated using DFT methods. The results of these calculations agree qualitatively well with the experimental results.
Optically active amines, used as racemic mixture, also replace triethylamine in the chiral cavity of 1–. Due to diastereotopic interactions between the host and guest an unequivocal assignment of the 1H NMR signals for the corresponding enantiomers of amines is possible. Thus, the zirconate anion 1– can be used for the chiral recognition.
1H NMR studies have further demonstrated that the binding energy for the (S)-enantiomer of 1-phenylethylamine with the pocket of 1– is larger than for the (R)-enantiomer, which could be confirmed by DFT calculations. This result allows the preparative application of chiral recognition, as shown by amidation of a racemic mixture of an amine with pivaloyl chloride. In this experiment an enantiomeric excess of 33% was found for the endohedral linked (S)-enantiomer.
An attempt to activate enamines as dienophiles for Diels-Alder reaction via protonation of the nitrogen atom in the chiral pocket of 1– leads to the formation of an embedded iminium cations, which could be isolated as a host-guest compound. This result contrasts the outcome of the protonation of enamines without the pocket of 1–, which illustrates an equilibrium between free enamine and triethylammonium chloride on the one side and the iminium cation and triethylamine on the other side. By evaporation of the solution, which contains the enamine as well as the iminium cation without 1–, the enamine is recovered quantitatively and no solid iminium salt could be observed.
The formation of iminium cations within the chiral pocket of 1– opens a way to an enantio-selective nucleophilic addition to the a-carbon atom of iminium cation. While the additions of carbon nucleophiles like malonate or benzoylacetonate were not successful, the addition of cyanide, with either triethylammonium chloride or the complex [Et3NH]1, produced in very good yield a-aminonitriles, which are potential precursors for a-amino acids.
A stereochemical influence of the zirconate anion 1– on the formation of a-aminonitriles was observed for 1-(2-phenyl-1-propenyl)-pyrrolidine, which after the transformation to a-amino-nitrile contains two stereogenic centers and exists as an erythro and threo diastereomer. The modified Strecker reaction increases the ratio of the threo to the erythro diastereomer by a factor of 7.5 in the presence of the zirconate complex [Et3NH]1 compared to the reaction with HNEt3Cl. This effect is obviously a consequence of the stereochemical induction of the chiral cavity of 1–.
The investigation of the properties of the chiral pocket 1– not only reveal the easy exchange of the endohedral linked triethylamine but also the direct synthesis of 1– by other amines and enamines as a trap for produced hydrogen chloride. The formed supramolecular complexes contain the new amines and enamines as ammonium or iminium cations encapsulated in the pocket of 1–. Since the chiral pocket of 1– recognizes enantiomers of optically active amines, the synthesis of 1– in the presence of a racemic mixture of a chiral amine results in a preferential inclusion of one enantiomer in the cavity of 1–. Using this procedure with 1-phenylethylamine, 65% enantiomeric excess of (S)-enantiomer of the amine was observed, which is endohedrally bound in the chiral pocket of 1–.
