Abstract: Die massive, transiente Produktion von reaktiven Sauerstoffspezies (oxidative burst) ist eine wichtige Komponente der pflanzlichen Pathogenabwehr. Die molekularen Mechanismen, die zur Entstehung des oxidative burst in Pflanzen führen, sind noch nicht aufgeklärt. Es wurden zwei cDNA Klone, rboh1 und rboh2 (respiratory burst oxidase homolog) isoliert, deren abgeleitete Aminosäuresequenzen signifikante Homologien zur katalytischen Untereinheit der NADPH-Oxidase aus Säuger-Phagozyten, GP91phox, aufweisen. In Phagozyten wird die Produktion von Superoxidanionen (respiratory burst) durch einen multimeren NADPH-Oxidase-Komplex katalysiert und dient der direkten Bekämpfung eingedrungener Erreger. Durch heterologe Expression der RBOH-Proteine in der Hefe Saccharomyces cerevisiae konnte in den mikrosomalen Fraktionen der Hefen NAD(P)H-abhängige O2--Produktion detektiert werden. Es wurde gezeigt, dass die O2--Bildung in RBOH2-Hefemikrosomen unabhängig von der Anwesenheit von Ca2+ war. Hingegen wurde in RBOH1-Hefemikrosomen unter Verwendung eines Ca2+-haltigen Extraktionspuffers Enzymaktivität nachgewiesen. Übereinstimmend damit enthält die RBOH1-Aminosäuresequenz mindestens ein putatives Ca2+-bindendes EF-Hand-Motif. Die Enzymaktivität der rekombinanten RBOH-Proteine wurde durch den Flavin-Oxidase-Hemmstoff DPI inhibiert. Die Applikation ähnlicher Konzentration DPI hemmt in vielen Pflanze/Pathogen-Systemen die Produktion von ROS und nachfolgenden Abwehrreaktionen. In Petersiliezellkulturen wurde nach Behandlung mit einem pilzlichen oder bakteriellen Elicitor ein transienter 3 bis 5facher Anstieg der rboh2-Transkriptmenge mit einem Maximum nach 2 Stunden detektiert. Untersuchungen mit stabil transformierten rboh2-Sense-Zellkulturen lieferten erste Hinweise auf eine physiologische Rolle der RBOH-Proteine innerhalb des oxidative burst. Da keine rboh2-Antisense-Kulturen generiert werden konnten, in der die rboh2-Transkriptakkumulation vollständig unterdrückt war, sind weitere Experimente zur Funktionsaufklärung notwendig. In dieser Arbeit gelang es erstmals einen funktionalen Zusammenhang zwischen pflanzlichen Genen mit Homologie zu gp91phox und einer von vielen Arbeitsgruppen postulierten NADPH-Oxidaseaktivität der kodierenden Proteine herzustellen.
One of the earliest detectable pathogen responses and a crucial element of plant defense is the rapid and transient production of reactive oxygen species (ROS), primarily superoxide and hydrogen peroxide. To date the molecular mechanisms that are responsible for the production of ROS in plants are not known. We isolated two cDNAs rboh1 and rboh2 (respiratory burst oxidase homolog) from parsley encoding proteins with significant homologies to the catalytic subunit of the phagocytic NADPH oxidase, GP91phox. In phagocytic cells a multimeric NADPH oxidase complex generates microbicidal superoxide (respiratory burst) combating directly the pathogen challenge. Heterolog expression of the RBOH proteins in the yeast Saccharomyces cerevisiae results in detection of NAD(P)H-dependent O2- production in the microsomal yeast fractions expressing the recombinant proteins. It has been shown that the NADPH oxidase activity of RBOH2 was independent of Ca2+, whereas in RBOH1 yeast fractions O2- production was detectable only after addition of Ca2+ to the extraction buffer. This matches with the RBOH1 amino acid sequence which contains at least one putative calcium binding EF-hand motif. The Flavine oxidase inhibitor DPI blocked the NADPH oxidase activity of the recombinant proteins with a similar concentration that is necessary to compromise the induction of the oxidative burst in parsley and other plant systems. RT-PCR experiments revealed a 3 to 5 fold transient increase of the rboh2 transcript accumulation in response to treatment of cultured parsley cells with elicitors derived from fungi or bacteria. We used stable transformed suspension cultured parsley cells expressing either sense or antisense rboh2 cDNA and assayed for elicitorresponsiveness to elucidate a physiological role of the RBOH proteins. The characterization of two rboh2 sense lines provided first evidence for the involvement of the RBOH proteins in a signal transduction cascade triggering defense responses. Since no transgenic cell culture with completely suppressed rboh2 transcript accumulation could be generated further studies will be necessary to demonstrate a physiological function. For the first time a functional link between plant gp91phox homologs and a postulated NADPH oxidase activity of the encoding proteins could be demonstrated.
