Abstract: Gegenstand der vorliegenden Arbeit ist die Synthese neuartiger potentieller antiviraler- und antiprionen-aktiver Acridinverbindungen. Durch die Anwendung der Konzepte der Interkalation an DNA und der Erzeugung hoch reaktiver Hydroxylradikale durch die Fenton-Reaktion gelangt man zu antiviralen EDTA-Verbindungen der Grundstrukturen A und B, welche zur Behandlung von Blutpräparaten, sowie zur Desinfektion in Krankenhäusern verwendet werden könnten. Bei den bisher bekannten Wirkstoffen zur Behandlung von Prionenerkrankungen (BSE, vCJD, TSE)) steht die Verhinderung der Anreicherung fehlgefalteten Prionenproteins im Vordergrund. Aus wichtigen Vertretern dieser Verbindungen lassen sich die quinacrinanalogen Monoacridinverbindungen der Grundstruktur C und die Bis-Acridinverbindungen der Grundstruktur D ableiten. Als zentrale Intermediate dienen bei der Synthese der Zielverbindungen substituierte 9-Chloracridine, welche sich aus den entsprechenden Anthranilsäuren ableiten. Zur effektiven zielgerichteten Darstellung dieser Verbindungsklasse wurden durch die Variation der Reaktionsbedingungen zwei verschiedene übergangsmetall-katalysierte Synthesen etabliert werden. Es handelt sich hierbei um Kupfer(I)-katalysierte Ullman-Jourdan-Reaktionen sowie Palladium(0)-katalysierte Reaktionen nach Buchwald-Hartwig. Durch die Reaktion der auf diese Weise erhaltenen 9-Chloracridine mit monogeschützten 1,ω-Diaminoalkanen verschiedener Kettenlänge und anschließende Umsetzung mit EDTA-Triethylester oder EDTA-Dianhydrid konnten die antiviralen Verbindungen der Grundstrukturen A und B erhalten werden. Durch die Reaktion der 9-Chloracridine mit N,N-Diethyl-1,4-diaminopentan, ungeschützten 1,ω-Diaminoalkanen oder anderen Diaminoverbindungen konnten die antiprionen-aktiven Substanzen der Grundstrukturen C und D erhalten werden. Die antibiotische Wirksamkeit der Fenton-aktiven Verbindungen wurde mit Hilfe eines Bakteriophagenscreenings getestet. Hierbei konnte eine sehr hohe Aktivität der Bisinterkalatoren der Grundstruktur B festgestellt werden. Die höchste Aktivität wurde bei Verbindungen der Kettenlänge 6 gefunden. Die Antiprionenaktivität der Verbindungen der Grundstrukturen C und D wurde mit Hilfe der FACS-Analyse aggrigierter Mausprionenproteine untersucht. Hierbei wurde eine deutliche Abhängigkeit sowohl von der Struktur des Acridins als auch des Diaminospacers gefunden. Die höchsten gefundenen Aktivitäten waren vergleichbar mit denen der bisherigen Leitsubstanzen, welche durch Zelltests gefunden wurden.
The purpose of this work was the synthesis of novel potential antiviral and antiprion active acridine compounds. The basic structures A and B of the antiviral EDTA compounds were achieved by the application of the concepts of intercalation on DNA and the generation of highly reactive hydroxyl radicals by the Fenton reaction. This substances could be used for the treatment of blood preparations and for disinfection in hospitals. By the known agents for the treatment of prion diseases (BSE, vCJD, TSE) the prevention of the accumulation of the false folded prion protein takes center stage. The basic structures of the quinacrine analogous monoacridines C and the bis-acridines D were achieved from the leading substances in TSE treatment.By the synthesis of the target compounds the substituted 9-chloroacridines were key intermediates and usually achieved from anthranilic acids. For an effective and target-oriented manufacture of this class of compounds two different transition metal catalysed synthesis were established. Here its about the copper(I)-catalysed Ullmann-Jourdan reaction as well as the palladium(0)-catalysed Buchwald-Hartwig amination. By the reaction of the obtained 9-chloroacridines with mono protected 1,ω-diaminoalkanes with different chain lengths following an reaction with triethyl EDTA or EDTA dianhydride the antiviral active basic structures A and B were obtained. By the reaction of the 9-chloroacridines with N,N-diethyl-1,4-diaminopentane, unprotected 1,ω-diaminoalkanes or other diamino compounds the antiprion active basic structures C and D were obtained. The antibiotic activities of the Fenton-active compounds were tested with a bacteriophage screening assay. Here the very high activities were found for the bisintercalating compounds with the basic structure B. The highest activity was found for substances with the diaminoalkane chain length 6. The antiprion activity of the compounds with the basic structures C and D were tested by the FACS analysis of aggregate mouse prion protein. An explicit dependency on the structure of the acridine and of the diamino spacer was found. The highest activities found were comparable with the leading compounds found by cellular screening methods.
