Abstract: In dieser Arbeit wurden erstmals vier verschiedene Fluoreszenzproteine (FP) als Marker für in vivo Xenograftimaging vergleichend untersucht. Es wurde gezeigt, dass mit nicht-invasivem in vivo Multispektral-Fluoreszenzimaging eine Quantifizierung der Tumorlast in humanen Xenograftmodellen des kolorektalen Karzinoms in der athymischen Nacktmaus prinzipiell möglich ist. Im s. c. Modell sind FP des kurz- wie langwelligeren Spektralbereichs geeignet. Tiefrote FP erlauben das Monitoring von Bauchraumtumoren, wobei tieflokalisierte Tumore nur unzureichend erfasst werden. Dies schränkt die Verwendung dieser FP für das Monitoring von orthotopen oder systemischen Modellen erheblich ein. Eine modellabhängige Tumorakkumulation infrarotmarkierter HES konnte im s. c. Xenograft gezeigt werden. Das Kontrastmittel brachte zusätzliche in vivo Informationen zu tieflokalisierten Tumoren. Somit könnte HES in der Diagnostik wie als biokompatibler Arzneiträger tumortypabhängig eingesetzt werden.
For the first time, in this PhD thesis four different fluorescence proteins (FP) were examined with a comparative approach. It was shown that quantification of tumour burden in human xenograft models of colorectal carcinoma in athymic nude mice is possible using non-invasive in vivo multispectral fluorescence imaging. Short- and long-wave FP are applicable in s. c. models. Deepred FP enable monitoring of abdominal xenografts with an insufficient acquisition of deeply located tumours. This fact limits the usage of such FP for monitoring of orthotopic or systemic models extensively. The application of infrared marked HES resulted in a model depending tumour accumulation in s. c. xenografts. The macromolecular contrast agent yielded additional in vivo information for deeply located tumours. Thus, HES could be utilised in diagnostics and as biocompatible drug delivery system depending on the tumour type.
