Abstract: Einleitung: Am isoliert-perfundierten Rattenherzen wurden mit einem Pharmakokinetik-Pharmakodynamik Modell die Effekte pathophysiologischer Einflüsse auf die Bindungs- und Signaltransduktionsparameter der α1-Adrenozeptoren untersucht. Methoden: Den Herzen wurde kontinuierlich Phenylephrin zugeführt und [3H]-Prazosin als Einzeldosis über 1 min infundiert. Die simultane Analyse der zeitlichen Verläufe der Prazosinkonzentration sowie des inotropen Effekts unter Nutzung der Agonist-Antagonist-Interaktion und einer nicht-linearen Regression ermöglichte die Schätzung von Rezeptoraffinität und der Parameter des "operational model of agonism". Hypertrophie wurde durch Vorbehandlung mit Isoprenalin (5 mg.kg−1) für 7 Tage induziert. Ergebnisse: Partielle Hypoxie führte zur signifikanten Reduktion der maximal am System möglichen Wirkung. Bei Hypertrophie kam es zur signifikanten Abnahme der Rezeptordichte und einem Anstieg des maximal erreichbaren Effekts. Schlussfolgerung: Die PK/PD-Modellierung erweist sich als geeignet, Parameteränderungen der α1-Adrenozeptoren unter pathophysiologischen Einflüssen zu ermitteln.
Introduction: A pharmacokinetic-pharmacodynamic modeling approach was used to analyse the effects of different pathophysiological influences on α1-adrenoceptor binding and signal transduction in isolated rat hearts. Methods: In Langendorff-perfused rat hearts phenylephrine was continuously infused and [3H]-prazosin was injected as a single dose (infused over 1 min). Simultaneous analysis of the time courses of prazosin outflow concentration and inotropic response using an agonist-antagonist interaction model and non-linear regression allowed to estimate receptor affinity, as well as the parameters of the operational model of agonism. Hypertrophy was induced by a pretreatment with isoprenaline (5 mg.kg−1) for 7 days. Results: Mild hypoxia significantly reduced the maximum response achievable in the system. Hypertrophy led to a significant reduction in receptor density and an increase in the maximum response achievable in the system. Conclusions: The experiments showed the shifting of the α1-adrenoceptor system's parameters both at the receptor and post-receptor level as a result of different pathophysiological influences.
