Abstract: Tuberkulose ist eine der weltweit am meisten verbreitete Infektionskrankheit, der jährlich 1.3 Mio. Menschen zum Opfer fallen. Besorgniserregend ist der Anstieg multiresistenter Fälle, auch in Industrienationen. 2009 wurden die antimykobakteriellen 8-Nitro-1,3-benzothiazinone (BTZ) entdeckt. Ziel dieser Arbeit war die Evaluierung verschiedener Synthesewege zu BTZs, wobei eine neue vereinfachte 2-Stufen Synthese etabliert wurde. Verschiedene BTZ-Derivate wurden synthetisiert, an mehreren Mykobakterienarten getestet, sowie pharmakokinetische Eigenschaften wie Löslichkeit und metabolische Stabilität bestimmt. Aus den Ergebnissen wurden wichtige Struktur-Wirkungsbeziehungen der antimykobakteriellen BTZs abgeleitet. Durch bioisosteren Austausch des Schwefels wurde eine neue antimykobakterielle Substanzklasse entdeckt – die 8-Nitro-1,3-benzoxazinone (BOZ). Die Röntgenkristallstrukturanalyse der BOZ mit dem Targetenzym DprE1 zeigte den gleichen Bindungsmechanismus wie die BTZs, wodurch BOZs als neue antimykobakterielle Klasse etabliert wurden.
Tuberculosis is one of the most widespread infectious diseases and a leading cause of death worldwide. The emergence of multidrug and extensively drug resistant tuberculosis is a growing health problem. In 2009, 8-nitro-1,3-benzothiazinones (BTZ) were discovered as highly active antitubercular agents. Scope of this work was the evaluation of different synthetic pathways to BTZs, resulting in the development of a novel straightforward two-step synthesis. A variety of new BTZ derivatives were synthesized, tested against different mycobacteria species and pharmacokinetic properties such as solubility and microsomal stability were determined. Essential structure activity relationships for antitubercular BTZs were concluded. A second antimycobacterial scaffold was developed by replacing the sulfur of BTZs with its isoster oxygen – 8-nitro-1,3-benzoxazinones (BOZ). Confirmed by crystal structure with the target enzyme DprE1, BOZs share the same mechanism of action with BTZs and are established as novel antitubercular scaffold.
