Abstract: Adipokine sind Proteine, die vorrangig von Adipozyten sezerniert werden und eine wichtige Rolle bei peripheren und zentralen physiologischen Prozessen spielen. In der folgenden Arbeit wurde untersucht, ob Adipokine im männlichen Reproduktionstrakt nachweisbar sind und wie sie die Spermienparameter beeinflussen. Aus den Ergebnissen der Beobachtungsstudie geht hervor, dass Adiponektin und Progranulin des Seminalplasmas als positive Marker der Ejakulatqualität fungieren. In Spermatozoen und Testis wurden die Proteine Adiponektinrezeptor 1, Leptin, dessen Rezeptor und NAMPT nachgewiesen. Adiponektin und Leptin beeinflussten die Spermienmotilität und erhöhten die Tyrosinphosphorylierung von Proteinen, die an der Kapazitation beteiligt sind. NAMPT hingegen wird als ein Marker der Reifung humaner Spermatozoen vermutet. Zusätzlich wurde der Prozess der Autophagie untersucht. In humanen Spermatozoen und Testis waren die Proteine LC3B und Beclin-1 nachweisbar.
Adipokines, proteins predominately secreted by adipocytes, play major role in peripheral and central physiological processes. The aim of this study was to evaluate the presence of adipokines in human male reproductive tract and their impact on sperm function parameters. The results of the observational study suggested, that adiponectin and progranulin found in seminal plasma function as positive markers for fertility prognosis. We were able to detect adiponectin receptor 1, leptin and its receptor and NAMPT in spermatozoa and testis. Adiponectin and leptin affected sperm motility and increased the tyrosine phosphorylation of proteins involved in capacitation. However, NAMPT was suggested to be a maturation marker of human spermatozoa. Additionally, the process of autophagy was investigated. Autophagy related proteins LC3B and Beclin-1 were present in human spermatozoa and testis. Immature spermatozoa might be more susceptible to autophagy than their mature counterparts.
