Abstract: Die Alzheimer Demenz (AD) ist die häufigste neurodegenerative Erkrankung. Aufgrund der multifaktoriellen Pathogenese sind neue Antidementiva mit neuen Wirkmechanismen notwendig, da bisher zugelassene Wirkstoffe nur kurzzeitig eine symptomatische Verbesserung ermöglichen. Eine wichtige Ursache des Neuronenuntergangs ist die pathologische Veränderung des Tau-Proteins mit dem Verlust seiner zellulären Funktion. In diesem Zusammenhang konnten verschiedenen Proteinkinasen eine Beteiligung an der Entwicklung der AD nachgewiesen werden. Im Rahmen dieser Arbeit wurde eine Vielzahl strukturell variierter, ATP-kompetitiver Proteinkinaseinhibitoren mit 1-Aza-9-oxafluoren-Grundstruktur synthetisiert und eine breite Auswahl dieser in vitro auf ihre inhibitorische Wirkung an acht AD-relevanten Proteinkinasen (CDK1/B, CDK2/E, CDK4/D, CDK5/p25, GSK-3β, ERK2, JNK3 und p38-γ) untersucht. Desweiteren wurde der Bindungsmodus einiger Verbindungen in Docking-Studien näher betrachtet.
Alzheimer's disease (AD) is the most common neurodegenerative disease. Due to the multifactorial pathogenesis new anti-dementia drugs with novel mechanisms of action are necessary because licensed drugs only have a short-term symptomatic improvement. A major cause of neuronal death is the pathological change of tau protein and the loss of its cellular function. In this context various protein kinases have been discussed to make a contribution to the progression of AD. In this work a great number of structurally varied, ATP-competitive protein kinase inhibitors of the 1-aza-9-oxafluorene type were synthesized. A wide variety of compounds was tested in an in vitro assay for inhibitory activity against eight AD relevant protein kinases (CDK1/B, CDK2/E, CDK4/D, CDK5/p25, GSK-3β, ERK2, JNK3 and p38-γ). Furthermore, docking studies of some compounds were made to predict the preferred binding mode.
