Abstract: Das β-Proteobakterium Cupriavidus metallidurans CH34 ist aufgrund einer Vielzahl von Resistenzdeterminanten an schwermetallkontaminierte Lebensräume angepasst und ein Modellorganismus für Untersuchungen zur Metall- und im speziellen Zinkhomöostase. Als Co-Faktor einer Vielzahl von Metallo-Proteinen nimmt Zink eine bedeutende Rolle in biologischen Systemen ein, obwohl es in hohen Konzentrationen zu toxischen Effekten führt. In dieser Arbeit wurde die Zugehörigkeit von drei putativen als G3E P-loop GTPasen annotierten Metall-Chaperonen CobW1/2/3 auf die Zinkhomöostase, in Zusammenhang mit der Zinkspeicherung und Übergabe untersucht. Die Zugehörigkeit der für die Chaperone kodierenden Gene zum Zur-Regulon stellt eine Verknüpfung zum Zinkhaushalt von C. metallidurans her. Ebenso die Metallbindekapazitäten der Proteine, die aufgrund unterschiedlicher Sekundärstrukturen und Regulation vermutlich unterschiedliche Rollen in der Zinkhomöostase von C. metallidurans einnehmen.
The β-proteobacterium Cupriavidus metallidurans CH34 is adapted to heavy metal contaminated habitats due to a large number of resistance determinants and is a model organism for investigations into metal and in particular zinc homeostasis. As a co-factor of a large number of metalloproteins, zinc plays an important role in biological systems, although it leads to toxic effects in high concentrations. In this thesis the association of three putative metal chaperones CobW1/2/3 annotated as G3E P-loop GTPases to zinc homeostasis was investigated in connection with zinc storage and transfer. The affiliation of the genes encoding the chaperones to the regulon establishes a link to the zinc homeostasis of C. metallidurans. The same applies to the metal binding capacities of the proteins, which probably play different roles in the zinc homeostasis of C. metallidurans due to different secondary structures and regulation.
