Abstract: Der filamentöse Ascomycet Colletotrichum graminicola ist der Erreger der Anthraknose Blattflecken und –stängelfäule an Mais. Epigenetische Regulationsmechanismen für die Bildung von Heterochromatin, wie H3K9 Trimethylierung (H3K9me3) oder DNA-Methylierung waren in diesem Organismus bislang wenig verstanden. Gezielte Knockout-Analysen von putativen Histon- und DNA-Methyltransferasegenen decken auf, dass diese Faktoren eine entscheidende Rolle in der Heterochromatisierung von C. graminicola spielen. Das Heterochromatin Protein 1 (HP1) fungiert dabei als Adapter zwischen H3K9me3 und der DNA-Methylierung. Weiterhin wurde gezeigt, dass dieses Heterochromatin Protein 1 essentiell für die Pathogenität des Pilzes ist.
The filamentous ascomycete Colletotrichum graminicola is the causative agent of anthracnose leaf spot and stalk rot on corn. Epigenetic regulatory mechanisms for the formation of heterochromatin, such as H3K9 trimethylation (H3K9me3) or DNA methylation, have been poorly understood in this organism. Targeted knockout analyzes of putative histone and DNA methyltransferase genes reveal that these factors play a crucial role in the formation of heterochromatin in C. graminicola. The heterochromatin protein 1 (HP1) acts as an adapter between H3K9me3 and DNA methylation. Furthermore, it has been shown that this heterochromatin protein 1 is essential for the pathogenicity of the fungus.
