Abstract: Propranolol HCl Pellets wurden mit Polymermischungen aus Polyvinylacetat und Polyethylenglycol-Polyvinylalkohol überzogen. Im Rahmen der vorliegen Arbeit wurden der Einfluss der Formulierung und der Coatingbedingungen auf die Propranolol HCl Freisetzung aus PVAc/PVA-PEG überzogenen Pellets untersucht. Außerdem wurde die Lagerungsstabilität analysiert. Mittels ESEM konnten morphologische Veränderungen des Polymerfilms, die durch Curing oder Lagerungsbedingungen hervorgerufen wurden, untersucht werden. Ein weiterer Aspekt dieser Arbeit war es, den zugrundeliegenden Freisetzungsmechanismus PVAc/PVA-PEG überzogener Propranolol HCl Pellets, die osmotisch aktive Agenzien enthalten, zu untersuchen. Quellungsexperimente und NMR Relaxometrie zeigten einen schnellen Wassereinstrom durch die Polymermembran in die Pellets. Diese Ergebnisse wurden durch ESR Experimente bestätigt. Die Wirkstofffreisetzung wird durch die Coatingzusammensetzung sowie durch osmotische Substanzen im Pelletkern gesteuert.
Propranolol HCl pellets were coated with polymer blends of poly(vinyl acetate) and poly(vinyl alcohol)-poly(ethylene glycol) copolymer. Within the present thesis the impact of formulation modifications and coating conditions on the Propranolol HCl release from PVAc/PVA-PEG coated pellets were investigated. In addition, stability issues were examined. ESEM experiments allowed to analyze morphological changes of the polymer film coat induced by curing or storage conditions. Another main aspect of this work was to elucidate the underlying drug release mechanism of PVAc/PVA-PEG coated Propranolol HCl pellets comprising of osmotically active ingredients. Swelling experiments and NMR relaxometry revealed a fast water diffusion through the polymer membrane into the pellet cores. These results were underlined by EPR experiments. The drug liberation was found to be controlled by the coating composition and by osmotically active ingredients within the pellet core.
