Abstract

Die steigende Inzidenz von Wundinfektionen bei MRSA führt zu hoher Morbidität und Mortalität. In dieser Studie sollte untersucht werden, ob die antimikrobielle Wirksamkeit von topisch appliziertem Moxifloxacin im Vergleich zu einem Trägerkontrollengel, Mupirocin, Linezolid und Gentamicin, unter Zuhilfenahme eines porcinen Infektions- und Ratten-Verbrennungsmodell gegeben ist. Nach Wundsetzung wurden die Tiere mit 1 x 108 CFU von MRSA oder P. aeruginosa inokuliert und therapiert. Die Ergebnisse aller topisch applizierten Antibiotika beweisen den gewünschten Effekt einer signifikanten Bakterienreduktion und erreichten Werte von < 105 CFU/g Gewebe. Zusätzlich konnte in der histologischen Analyse ein erhöhtes Wundheilungspotential von Moxifloxacin beobachtet werden. Die vorliegenden Ergebnisse zeigen, dass die topische Therapie mit Moxifloxacin eine neue Therapieoption darstellt.

Efficacy of topically delivered moxifloxacin against wound infection by Pseudomonas aeruginosa and methicillin-resistant Staphylococcus aureus

Wound infection is a common risk for patients with chronic nonhealing wounds, causing high morbidity and mortality. In this study, topical application of moxifloxacin was investigated in comparison to mupirocin, linezolid, and gentamicin using a porcine wound infection and a rat burn infection model. Both animal models were performed either by an inoculation with MRSA or P. aeruginosa. Wound fluid and tissue samples were taken for bacterial counts. All antibiotics showed efficient reduction of bacterial counts, and except with MRSA, infected burn wounds reached bacterial counts below 105 CFU/g tissue. Additionally, moxifloxacin was observed to promote wound healing as determined by histologic analysis. Moxifloxacin is therefore an ideal candidate, due to its broad antibacterial spectrum, its high efficiency, and its potential to promote wound healing.
