Mikrofluidisches System mit integrierter Multisensorik für mehrdimensionale Screenings in der miniaturisierten Umwelttoxikologie

One of the elementary goals of human health care and sustainable environmental management is protecting organisms from the damaging effects of toxic substances. Hazard assessments often rely on means of quantifying effects caused by a single substance, rather than by assessing the effects of several chemicals present at the same time. In reality, however, organisms are exposed to a complex and always fluctuating mixture of substances. Experimental investigations become exponentially more complex due to dimensionality. Thus, that each additional involved substance requires a tremendous amount of further investigation. Conventional toxicological methods cannot adequately address this challenge. New techniques are therefore required for allowing investigations with larger sets of samples, with lower consumption of chemicals and biological materials, and within a reasonable timeframe. Such a possibility is offered by the application of microfluidics. The aim of this work is to develop a compact, automated fluid-handling and measurement system with integrated multi-sensors utilizing the segmented flow method in order to characterize highly-resolved dose response relationships for single- and combined effects of environmental harmful substances.With the developed lab- and compact systems, it is possible to generate a microfluid segment series for the determination of two, three and five-dimensional concentration fields. Successful investigations of different combinations of substances were performed on Escherichia coli, leukemia cells IPC-81, Streptomyces tendae F4, Streptomyces acidiscabies E13, Psychrobacillus psychrodurans UrPLO1 and the green algae Chlorella vulgaris.Through the integration of pO2-sensitive nanobeads inside microfluid segments, a simultaneous, non-invasive readout of the segment internal pO2-development during the time and concentration dependent cultivation process of the heavy metal-tolerant bacteria Streptomyces acidiscabies E13 and Psychrobacillus psychrodurans UrPLO1 inside microfluid segments was achieved.Using this approach a rapid analysis of interferences among drugs of the substance classes phenols, ionic liquid, heavy metal ions, antibiotics, herbicides, fungicides, drugs based on ACE or CSE inhibitors, food ingredients and metal nanoparticles can be performed as well as the fast and precise determination of the combind substance toxicity. Therefore, the end points of growth, autofluorescence signal, and the segment-internal pO2-change were obtained via a two-channel micro flow-through fluorimeter and a four-channel micro flow-through photometer.From both of the highly-resolved dose-response relation of single substances and the multi dimensional screenings, various unexpected effects were found. The results have demonstrated the applicability of the developed compact system and the huge potential of the droplet-based microfluidic technique for miniaturized screenings in environmental toxicology and drug development as well as related fields.

Die globale Belastung der Umwelt mit Schadstoffen stellt ein Problem sowohl für Mensch als auch für Ökosysteme dar. Schadstoffe kommen in der Umwelt selten einzeln, sondern meist in komplexen Gemischen vor. Deshalb nimmt die Diskussion über die Kombinationswirkung von Schadstoffen einen immer größeren Stellenwert ein. Die Größe des mehrdimensionalen Parameterfeldes erschwert die Anwendung toxikologischer Standardmethoden und macht die Einführungen neuer Screeningmethoden notwendig. Der Einsatz der Mikrosystemtechnik im Bereich der Biotechnologie bietet erhebliche Vorteile bezüglich Miniaturisierung und Automatisierung verglichen mit herkömmlichen Verfahren. Das Ziel des Promotionsvorhabens bestand in der Entwicklung eines kompakten, automatisierten Fluidhandling- und Messsystems mit integrierter Multisensorik unter Anwendung des Prinzips mikrosegmentierter Flüsse zur Charakterisierung hochaufgelöster Dosis-Wirkungs-Beziehungen für die Einzel- sowie Kombinationswirkung von Umweltschadstoffen.Mit den entwickelten Labor- und Kompaktsystemen konnten zwei-, drei- und fünfdimensionale Konzentrationsfelder in Mikrofluidsegmenten erzeugt werden. Die Wirkungen verschiedener Schadstoffkombinationen auf Darmbakterium Escherichia coli, Grünalge Chlorella vulgaris, Bodenbakterien Streptomyces tendae F4, Streptomyces acidiscabies E13, Psychrobacillus psychrodurans UrPLO1 und Vertebraten-Zelllinien IPC-81 wurden erfolgreich untersucht. Durch die Integration von pO2-sensitiven Nanobeads in die Mikrofluidsegmente gelang die interaktionsfreie Auslese des segmentinternen pO2-Wertes während der Zeit- sowie konzentrationsabhängigen Kultivierung der schwermetalltoleranten Bakterienstämmen Streptomyces acidiscabies E13 und Psychrobacillus psychrodurans UrPLO1 in Mikrofluidsegmenten.Mit den entwickelten Systemen konnten zahlreiche binäre- und ternäre Gemische aus verschiedenen Organismen modellhaft untersucht werden. Es konnten Datensätze zu hochaufgelösten Einzelwirkungs- und Kombinationsscreenings der Verbindungsklassen Phenole, ionische Flüssigkeiten, Schwermetalle, Antibiotika, Herbizide, Fungizide, Medikamente (ACE Hemmer und CSE Hemmer), Ernährungsbestandteile und Metallnanopartikel anhand der Endpunkte Wachstum, Autofluoreszenz sowie der Änderung der Lumineszenzintensität von pO2 sensitiven Nanobeads mittels eines 2 Kanal Mikrodurchflussfluorimeters und eines 4 Kanal Mikrodurchflussphotometers gewonnen werden. Sowohl aus hochaufgelösten Einzelwirkungsscreenings als auch aus mehrdimensionalen Screenings der Mikrofluidsegmente konnten neue Erkenntnisse gewonnen werden. Die Versuchsergebnisse demonstrieren bereits die Einsatzfähigkeit der Kompaktanordnung und das große Potential der tropfenbasierten Mikrofluidik für toxikologische Multiparameter-Screenings. Das entwickelte System bildet eine wichtige Grundlage für ein marktfähiges Kompaktgerät in der miniaturisierten Ökotoxikologie.
