Die Inzidenz von hellem Hautkrebs hat in den letzten Jahrzehnten, auch aufgrund zu hoher UV-Licht-Exposition, stark zugenommen. Die derzeitigen topischen Therapieoptionen sind entweder schmerzhaft und/oder weisen unzureichende Heilungsraten auf. Daher war es ein Ziel dieser Arbeit, einen neuen therapeutischen Ansatz für die topische Therapie der Aktinischen Keratose, des Plattenepithelkarzinoms und des Basalioms zu entwickeln. Die Pol α ist ein vielversprechendes Target, da sie die ersten Schritte der DNA Replikation katalysiert und mit einer Primase-Funktion assoziiert ist. Die 3-dimensionale Struktur des aktiven Zentrums der humanen Pol α konnte mittels Molecular Modelling konstruiert und eine Reihe bekannter Pol-α-Inhibitoren in das aktive Zentrum gedockt werden. Gemäß diesen Berechnungen wurden die neuen nukleosidischen Verbindungen HM-1-Oxim und isoHex-OH sowie die neuartigen Nukleotid-Analoga OxBu, OxIsohex, OxHex und deren Diethylester OxBu-DE und OxHex-DE identifiziert, synthetisiert und im Rahmen dieser Arbeit mittels in- vitro-Zelltests pharmakologisch gescreent. Mit HM-1-Oxim und isoHex-OH gelang es weder, den derzeitigen Standard der topischen Therapie – 5-FU – in seiner Wirkung zu übertreffen, noch Tumorzellen selektiv anzugreifen. Die Derivatisierung zum Phosphonat an der endständigen 5’-OH-Gruppe des Zucker- analogen Molekülteils führte zu den besser aktivierbaren Monophosphat-Analoga OxBu, OxIsohex und OxHex. Untersuchungen an normalen humanen Keratinozyten und an der Plattenepithelkarzinomzelllinie SCC25 ergaben eine deutliche Tumorselektivität, wobei OxBu und OxHex SCC25 Zellen schon im nanomolaren Bereich angriffen. Damit sind diese Phosphonate 1000- bzw. 10000-fach aktiver als die Vergleichssubstanzen Aphidicolin und 5-FU bzw. BuP-OH. Das Ziel, die Wirksamkeit von 5-FU und BuP-OH zu übertreffen, wurde erreicht und zudem tumorselektive Substanzen generiert. Aufgrund ihrer Zytotoxizität wurden OxBu und OxHex zusätzlich an den Tumorzelllinien HT29, MCF7, T24 und SISO untersucht und wirkten auch an diesen zuverlässig. Ferner wurden die Diethylester (DE) von OxBu und OxHex an SCC25 Zellen und NHK auf Zytotoxizität geprüft, weil die erhöhte Lipophilie zu einer verstärkten Aufnahme in die Targetzellen führen könnte, in welchen nach enzymatischer Esterspaltung die Muttersubstanzen freigesetzt werden. OxBu-DE und OxHex-DE zeigten allerdings ein schlechteres Wirkprofil als die Muttersubstanzen. Im zweiten Teil der Arbeit sollten eine analytische Detektionsmethode (HPLC) für die Pol-α-Hemmer und deren voraussichtliche Metaboliten entwickelt sowie eine Festphasenextraktionsmethode zur Isolierung dieser Metaboliten etabliert werden. Eine HPLC-Trennung der Modell-Nukleoside Thymidin und Guanosin inklusive deren DNA Basen und Nukleotide gelang mit Basislinientrennung und Detektion im nanomolaren Bereich. Mit dieser Methode ließen sich auch die Testsubstanzen BuP-OH, HM-1, HM-1-Oxim sowie die Nukleosid-Analoga Zalcitabin, Capecitabin, 5-FU und Aciclovir detektieren. Für die Phosphonate OxBu und OxHex konnte eine HPLC-Methode mit Adefovir als internem Standard erarbeitet werden, mit welcher auch die Detektion der (bisher noch nicht synthetisierten) Diphosphate der Testsubstanzen möglich scheint.
Due to an increased UV exposure, the number of patients with actinic keratosis and cutaneous squamous cell carcinoma has increased dramatically. Current therapy of actinic keratosis is either painful and/or cure rates are not totally satisfying. Therefore the aim of this study was an alternative approach like interference with DNA synthesis. The human DNA polymerase α catalyzes the very first steps in DNA replication and is tightly associated with a primase. Therefore, it is an interesting target. Recently, the three- dimensional structure of the active site of human DNA polymerase α was proposed based on the application of molecular modelling methods and molecular dynamic simulations. The modelled structure of the enzyme was used for docking several known inhibitors in its active site. Novel nucleosides as HM-1-oxime and isoHex-OH as well as the nucleotide analogs OxBu and OxHex and their diethylester prodrugs were identified, synthesized and subjected to an in- vitro-screening. Neither HM-1-oxime nor isoHex-OH surmounted the effects of 5-FU – the current standard for topical therapy of actinic keratosis. Moreover, no selectivity for tumor cells was observed. Costimulating the cells with 5-FU and the indicated agents did not improve the outcome either. Next, the phosphonate derivatives OxBu, OxIsohex and OxHex, containing a metabolically stable P-C bond, were developed in order to circumvent the initial activation step. Cytotoxicity testing in normal human keratinocytes (NHK) and SCC25 cells showed a distinct selectivity of the phosphonates for tumor cells. OxBu and OxHex affected SCC25 cells even in the nanomolecular range. Referring to the IC50 values, OxBu and OxHex exceeded the activity of 5-FU and aphidicolin about 1000fold, BuP-OH was outranged about 10.000fold. Summing up, the aim of exceeding the effects of 5-FU and BuP-OH was achieved. Additionally, the lack of tumor selectivity has been overcome. Due to their great potential as antitumor agents for actinic keratosis and squamous cell carcinoma OxBu and OxHex were tested in other tumor cell lines like HT29, MCF7, T24 and SISO cells, too. In fact, the phosphonates once more suppressed the viability of these tumor cell lines very efficiently. Since normal human keratinocytes produce great amounts of esterases the ability to affect NHK and SCC25 cells was investigated with the respective diethylesters of OxBu and OxHex, too. The higher lipophilicity may favour penetration of the target cell. Unexpectedly, the activity of the phosphonate diethylesters was less favourable as compared to the activity of OxBu and OxHex. Secondly, an analytical method (HPLC) ought to be investigated to detect the pol α inhibitors and their expected metabolites. Additionally, a solid phase extraction method ought to be established to extract these metabolites from the cells. An HPLC method was developed analysing the model compounds thymidine and guanosine including their respective DNA bases and nucleotides. Baseline separation and detection in the nanomolecular range was achieved. This method was applicable to the test compounds BuP-OH, HM-1, HM-1-oxime as well as to the nucleotide analogs zalcitabine, capecitabine, 5-FU and acyclovir. Aiming the detection of OxBu and OxHex likewise, an HPLC method usinig adefovir as internal standard was established. This method seems to be applicable for future detection of the potential phosphorylated metabolites of OxBu and OxHex.
