Simultane kombinierte TMZ/SU führt zu einer verbesserten Tumorkontrolle durch additive antiangiogene und synergistische proapoptotische Effekte. Die gesteigerte therapeutische Effektivität von kombinierter TMZ/SU-Therapie wird nur durch kontinuierliche gemeinsame Applikation, nicht jedoch durch sequenzielle Applikation beider Substanzen erreicht. Dennoch aggraviert diese Form der Kombinationstherapie Perizyten-vermittelte vaskuläre Resistenzmechanismen mit Expressionsveränderungen in den Signalwegen des Ang-1/Tie-2 und Dll-4- Systems.
Continuous application of combined TMZ/SU yielded beneficial therapeutic efficacy by additive antiangiogenic and synergistic pro-apoptotic effects. Sequential application of TMZ and SU in the angiogenic window does not add antitumor efficacy to monotherapy. Thus, long-term efficacy of combination therapy may be hampered by aggravated pericyte-mediated vascular resistance mechanisms with alterations in Ang-1-Tie-2 and Dll4/Notch pathways.
