Abstract:

Seitdem die Funktion von Kohlenhydraten als vielseitige Mediatoren von Zellerkennungsprozessen entdeckt wurde, ist das Interesse an dieser Stoffklasse wieder sprunghaft angestiegen. Besonders die zunehmende Kenntnis über die Bedeutung sialinsäurehaltiger Glycokonjugate bei zahlreichen physiologischen und pathologisch relevaten Erkennungsprozessen hat den Bedarf an synthetischen Sialinsäuren, Oligosacchariden und Glycokonjugaten für systematische biologische Studien enorm gesteigert. Sialinsäuren können mit Hilfe von CMP-Neu5Ac-Synthetasen aktiviert werden und stehen dann in dieser Form den Sialyltransferasen als Substrate zur Verfügung. Im Rahmen dieser Arbeit sollte die Substrattoleranz der aus Neisseria meningitidis rekombinant gut zugänglichen CMP-Neu5Ac-Synthetase nun genauer untersucht werden. Grundvoraussetzung für die Evaluierung der Substrattoleranz dieses Enzyms war vorab die Synthese geeigneter, strukturell breit abgewandelter Sialinsäureanaloga. Die für die Synthese von C-9- bzw. C-8-Sialinsäure-Analoga bewährte Indium-vermittelte Allylierung von Aldosen konnte hierbei erstmals auf die Synthese von neuen C-6- respektive C-7- Sialinsäurestrukturen ausgeweitet werden, wobei durch den Einsatz eines aciden Lösungsmittelsystems die Indium-vermittelten Allylierungen vollständig diastereoselektiv geführt werden konnten. Ausgehend von Zimtaldehyd gelang mit hohen Ausbeuten die Synthese der chemisch sensitiven Glycerinaldehyd-Derivate, die Indium-vermittelt zu den entsprechenden C-6-Sialinsäure-Analoga umgesetzt werden konnten. Die für die Synthese neuer C-7-Sialinsäure-Analoga benötigten Erythrose-Derivate konnten in einer analogen Prozedur synthetisiert und mittels der bewährten Barbier-Reaktion in die entsprechenden Sialinsäure-Derivate überführt werden. Mit Hilfe dieser chemisch synthetisierten Sialinsäure-Bibliothek konnte abschließend die Substrattoleranz der neuen CMP-Neu5Ac-Synthetase genau bestimmt werden.

Since then the function of carbohydrates as versatile mediators of cell recognition processes was discovered, the interest in this compound class again precipitously rose. Particularly the increasing knowledge of the meaning of sialic acid-containing glycoconjugates with numerous physiological and pathological recognition processes increased the requirement of synthetic sialic acids, oligosaccharides and glycoconjugates for systematic biological studies enormously. Sialic acids can be activated by CMP-NeuAc synthetases and then been available in this form for sialyltransferases as substrates. In the context of this work the substrate tolerance of the recombinant CMP-NeuAc synthetase from Neisseria meningitidis should be examined now more exactly. Basic condition for the evaluation of the substrate tolerance of this enzyme was first the synthesis of suitable and structurally broadly modified sialic acids analogs. The proved indium-mediated allylation of unprotected aldoses for the synthesis of C-9 or C-8-sialic acid analogs could here for the first time on the synthesis of new C-6 respectively C-7 sialic acid analogs be expanded, whereby by the use of an acidic solvent system the indium-mediated allylation could be led completely diastereoselectiv. On the basis of cinnamaldehyde the synthesis succeeded to that chemically sensitive glyceraldehyde derivatives were obtained with high yields, which could be transferred by indium-mediated chain-extension to the appropriate C-6-sialic acid analogs. The erythrose derivatives needed for the synthesis of new C-7-sialic acid analogs could be synthesized in a similar procedure and be transfered by means of the proven Barbier-reaction into the appropriate sialic acid derivatives. With the help of this chemically synthesized sialic acid library finally the substrate tolerance of the new CMP-NeuAc synthetase could be determined exactly.
