Abstract:

Gasvesikel sind intrazelluläre, gasgefüllte Proteinkörperchen, die von H. salinarum PHH1 während des gesamten Wachstums gebildet werden. Die 14 p-gvp-Gene, deren Expression die Bildung der zitronenförmigen Gasvesikel hervorruft, sind auf dem Plasmid pHH1 in der sogenannten p-vac-Region lokalisiert. Innerhalb dieser Region sind die Gene p-gvpACNO, mit p-gvpA für das Hauptstrukturprotein pGvpA, in eine Richtung orientiert, während die Gene p-gvpDEFGHIJKLM genaufwärts von p-gvpA in entgegengesetzter Richtung liegen. Die Notwendigkeit einzelner gvp-Gene für die Gasvesikelbildung konnte in Haloferax volcanii-Transformanten bestimmt werden. Erste Experimente wiesen darauf hin, dass die Gene p-gvpA und p-gvpO essentiell für die Gasvesikelbildung sind, während die Gene p-gvpCN und p-gvpDE nicht benötigt werden. Die Phänotypen von Hf. volcanii-Transformanten, die die p-vac-Region enthielten bei der jeweils ein Gen aus der p-gvpFGHIJKLM-Region deletiert war, zeigten, dass die Gene p-gvpH und p-gvpI für die Bildung der Gasvesikel nicht benötigt werden, während die Gene p-gvpF, p-gvpG, p-gvpJ, p-gvpK, p-gvpL und p-gvpM essentiell sind. Diese Ergebnisse zeigten, dass die acht Gene p-gvpA, p-gvpO, p-gvpFG und p-gvpJKLM der p-vac-Region für die Bildung von Gasvesikeln essentiell sind. Transformanten, die das Konstrukt p-gvpFGJKLM-AO enthielten, bildeten jedoch keine Gasvesikel wegen unzureichender Expression der Gene p-gvpJKLM. Die zusätzliche Überexpression der Gene p-gvpJKLM auf einem zweiten Vektorkonstrukt führte zu Transformanten, die Gasvesikel bildeten, was zeigte, dass die acht Gene p-gvpFGJKLM-AO zur Gasvesikelbildung ausreichen. Die Expression der 14 p-gvp-Gene geht von den Promotoren pA, pD, pF und pO aus, die vor den jeweiligen p-gvp-Genen liegen, zu unterschiedlichen Zeitpunkten des Wachstums aktiv sind und teilweise durch GvpD oder GvpE reguliert werden. Der pA-Promotor ist konstitutiv aktiv. Transkriptionsanalysen und Studien mit Hilfe des bgaH-Reportergens, das für eine halobakterielle b-Galaktosidase kodiert, zeigten, dass die Aktivität des pA-Promotors durch den Transkriptionsaktivator pGvpE erhöht wird. Ein repressorischer Effekt von pGvpD auf den pA-Promotor wurde nur in Kombination mit pGvpE beobachtet. Der pD-Promotor ist nur während der stationären Wachstumsphase aktiv. Transkriptionsanalysen und Untersuchungen mit dem bgaH-Reportergen zeigten, dass der pD-Promotor ebenfalls durch den Transkriptionsaktivator pGvpE aktiviert wird. Der pF-Promotor, der nur während des exponentiellen Wachstums aktiv ist und zur Bildung des p-gvpFGHIJKLM-Transkripts führt, wird weder durch pGvpD noch durch pGvpE reguliert. Ähnliches konnte für den ebenfalls exponentiell aktiven pO-Promotor gezeigt werden, der weder durch pGvpD noch durch pGvpE oder durch andere Genprodukte der p-vac-Region in seiner Aktivität beeinflusst wird.

Gas vesicles are intracellular, gas-filled, proteinaceous particles formed by H. salinarum PHH1 to provide buoyancy in the watery environment. The 14 different p-gvp-genes are present in the p-vac-region on plasmid pHH1 and the expression of these genes leads to spindle-shaped gas vesicles throughout growth. These genes are arranged as p-gvpACNO and, upstream of p-gvpA and oriented in the opposite direction p-gvpDEFGHIJKLM. The necessity of single gvp-genes for gas vesicle formation was analyzed in Haloferax volcanii transformants. Initial experiments indicated, that p-gvpA and p-gvpO are necessary for gas vesicle formation whereas p-gvpCN and p-gvpDE are not essential. The gas vesicle negative (Vac-) phenotypes of Hf. volcanii transformants harbouring the p-vac region with a deletion of a single gene in the p-gvpFGHIJKLM cluster indicated, that p-gvpH and p-gvpI are not essential for gas vesicle formation whereas the genes p-gvpF, p-gvpG, p-gvpJ, p-gvpK, p-gvpL and p-gvpM are required. These results implied that the eight genes p-gvpA, p-gvpO, p-gvpFG and p-gvpJKLM constitute the minimal number of genes required for gas vesicle formation. However, a transformant containing the construct p-gvpFGJKLM-AO did not produce gas vesicles due to unsufficient expression of the p-gvpJKLM-genes. Additional copies of the p-gvpJKLM genes resulted in transformants containing gas vesicles, suggesting that the eight genes p-gvpFGJKLM-AO represent indeed the minimal number of genes required for gas vesicle formation. The expression of the p-vac region is driven by the four promoters pA, pD, pF and pO located in front of the respective p-gvp gene. These promoters are active at different phases of growth and in part regulated by GvpD and GvpE. The pA-promoter is constitutively active. Transcript analyses and analyses using the bgaH reporter gene, encoding a halobacterial b-galactosidase, indicated that the pA-promoter is activated by pGvpE, a transcripional activator. A repressor effect of pGvpD was only observed together with GvpE. The pD-promoter is active only in stationary growth phase. Transkript analyses and analyses with the bgaH reporter gene indicated, that the pD-promoter is although activated by the transcripional activator pGvpE. The pF-promoter is only active during exponential growth and leads to the p-gvpFGHIJKLM transcript and neither pGvpE nor GvpD has effect on the pF-promoter activity. Similar results were obtained with the p-gvpO promoter which only active during exponential growth phase and is regluated neither by pGvpD nor by pGvpE or other gene products of the p-vac-region.
