Abstract: In der vorliegenden Habilitationsarbeit sollte die Rolle des Phospholipid-Transferproteins (PLTP), welches unter anderem den Transfer von Phospholipiden zwischen Lipoproteinen vermittelt, im Krankheitsbild der Atherosklerose untersucht werden. Die Evaluierung des PLTP erfolgte in klinischen Studien und zusätzlich in experimentellen Untersuchungen an verschiedenen Mausmodellen. In der vorliegenden Arbeit konnte gezeigt werden, dass 1. Eine Erhöhung der PLTP-Aktivität mit dem Vorliegen atherosklerotischer Erkrankungen assoziiert ist. 2. Inflammatorische Effekte des PLTP und die Assoziation zu den postprandial erhöhten Lipoproteinen Aspekte seiner potentiell pro-atherogenen Wirkung sind. 3. Die PLTP-Aktivität nicht mit der Prognose von Patienten mit terminaler Niereninsuffizienz oder koronarer Herzerkrankung assoziiert ist. Die prognostische Relevanz dieses Enzyms für Patienten mit atherosklerotischen Krankheitsbildern in Hinblick auf eine potentielle therapeutische Beeinflussung gilt es in weiteren Populationen zu prüfen.
In this postdoctoral lecture qualification the role of phospholipid-transferprotein (PLTP), which is mainly involved in the lipoprotein transfer of phospholipids, in atherosclerotic diseases is evaluated in clinical studies as well as in in vivo (mouse models) and in vitro experiments. It could be shown that first, an elevation of PLTP-activity is associated with atherosclerotic diseases, second, aspects of this potentially pro-atherogenic effects are associated with inflammation and elevation of postprandial lipoproteins, and third, PLTP-activity is not a prognostic marker for patients with atherosclerotic diseases. The prognostic impact of PLTP in patients with atherosclerotic diseases should be further evaluated as a potentially therapeutic target.
