Zusammenfassung

The localisation of the pathogenic prion protein PrPSc was investigated with light and electron microscopy. The PrPSc specific antibody 15B3 was tested for its efficiency in immuncytochemistry. Subsequently, an appropriate method was found to stain PrPSc selectively. PrPSc was detected in clathrin coated pits, early endosomes, late endosomes/lysosomes nad exosomes. PrPSc could not be observed in lipid droplets.
In the next part of the thesis different mutants of the prion protein carrying micro deletions within the hydrophobic region of the prion protein PrPC were investigated. The deletion mutants were checked for their convertibility into PrPSc in scrapie-infected neuroblastoma cells.

Zusammenfassung in einer weiteren Sprache

Die Lokalisation des pathogenen Prionproteins PrPSc wurde mit Hilfe von Licht- und Elektronenmikroskopie untersucht. Zum einen wurde der PrPSc spezifische Antikörper 15B3 auf seine Wirksamkeit in der Immunzytochemie getestet. Desweiteren wurde eine geeignete Methode gefunden, PrPSc selektiv anzufärben. PrPSc konnte in clathrin coated pits, frühen Endosomen, späten Endosomen/Lysosomen und Exosomen gefunden werden. PrPSc konnte nicht in Lipidkörpern gefunden werden.
Im nächsten Teil der Arbeit wurden verschiedene Prionproteinmutanten untersucht, die Mikrodeletionen in der hydrophoben Region des Prionproteins PrPC tragen. Diese Deletionsmutanten wurden in scrapie-infizierten Neuroblastoma Zellen auf ihre Konvertierbarkeit in PrPSc untersucht.
