Zusammenfassung

The selective synthesis of alpha and beta carbohydrate carbonates is reported. By reaction of these carbonates with amino-functionalised scaffolds, multivalent lectin ligands were obtained and have been tested in vitro and in vivo.
Two Split-Mix libraries, consisting of 13500 cyclic glycopeptides, were obtained by combinatorial solid phase peptide synthesis. By screening of these libraries potent ligands for the lectin Viscum album agglutinin were identified using the Edman-degredation.
Selected ligands were resynthesized and tested in solution.

Zusammenfassung in einer weiteren Sprache

In der Dissertation ist die alpha und beta selektive Synthese verschiedener Kohlenhydratcarbonate beschrieben. Durch deren Reaktion mit aminofunktionalisierten Gerüststrukturen konnten multivalente Lektinliganden synthetisiert und in vitro wie in vivo untersucht werden.
Durch kombinatorische Festphasensynthese konnten zwei Split-Mix Bibliotheken bestehend aus 13500 cyclischen Glycopeptiden erhalten werden. Beim anschließenden Festphasen-Screening der Neoglycopetidbibliotheken konnten hochaffine Liganden für das Lectin Viscum album agglutinin mittels Edman-Sequnzierung identifiziert werden.
Ausgewählte Liganden wurden nachsynthetisiert und mit einem verbesserten Inhibitionsassay (Enzym Linked Immunosorbent Lectin Assay) in Lösung untersucht.
